Sichelzellkrankheit (SCD) Biochip': Towards a Simple and Reliable Way to Monitoring Sichel Cell Disease
Die „sichelförmige“ Anämie wurde erstmals 1910 in den USA klinisch beschrieben, und das mutierte vererbbare Sichelhämoglobinmolekül wurde 1949 identifiziert. Die Pathophysiologie von SCD ist eine Folge der abnormalen Polymerisation von Sichelhämoglobin (HbS) und seiner Auswirkungen auf die Eigenschaften, Form und Dichte der Erythrozytenmembran und nachfolgende kritische Veränderungen der Funktion von Entzündungszellen und Endothelzellen. Unser Ziel ist es, die Auswirkungen von CMA-Anomalien bei SCD zu verstehen, indem wir eine Reihe anerkannter Wechselwirkungen bei einer Reihe klinischer Phänotypen untersuchen.
Bis heute bewegen sich korrelative Studien bei SCD von uns und anderen zwischen klinischen Berichten auf der Grundlage von Tests, Interventionen und Diagrammüberprüfungen von Einzelpersonen oder Gruppen von Einzelpersonen und im anderen Extrem der Identifizierung funktioneller Genpolymorphismen auf der Grundlage von Bevölkerungsstudien . Die Forscher möchten diese Studien durch eine systematische Untersuchung der Zellmembraneigenschaften und des Aktivierungsstatus ergänzen. Von den hämatologischen Störungen kann SCD ungewöhnlich anfällig für eine solche Untersuchung sein.

Einschlusskriterien
- Männlich oder weiblich ≥ 12 Jahre zum Zeitpunkt der Einwilligung (Einschreibung).
- Dokumentation Sichelzellanämie, einschließlich HbSS- oder Compound-heterozygus-HbSC- oder HbSβ-Thalassämie-Diagnose, nachgewiesen durch ein oder mehrere klinische Merkmale.
- Schriftliche Einverständniserklärung (und ggf. Zustimmung) des Probanden oder des gesetzlichen Vertreters des Probanden und Fähigkeit des Probanden, die Anforderungen der Studie zu erfüllen.
Ausschlusskriterien
- Vorhandensein eines Zustands oder einer Anomalie, die nach Ansicht des Prüfarztes die Sicherheit des Patienten oder die Qualität der Daten gefährden würde.

Entwicklung eines SCD-Biochips, mit dem wichtige zelluläre Eigenschaften und Wechselwirkungen untersucht werden können, einschließlich zellulärer, adhäsiver und entzündlicher Eigenschaften von RBC und WBC sowie Eigenschaften von zirkulierenden endothelialen und hämatopoetischen Vorläuferzellen.

Adhäsion roter und weißer Blutkörperchen an Biomoleküle (z.
Laminin, Fibronectin und Selectine) werden auf einem mikrofluidischen Gerät, dem SCD Biochip, gemessen.
Die Adhäsion wird unter normalen und sauerstoffarmen Bedingungen gemessen.
Adhäsionen werden in Bezug auf klinische Befunde wie hämatologische Parameter und Hinweise auf Vasookklusion, Schmerzen, Entzündung und Vaskulopathie bei Patienten mit Sichelzellanämie beurteilt.endothelial
und hämatopoetische Vorläuferzellen basierend auf Membraneigenschaften und Adhäsion.
