  • 환자는 췌장 선암종의 조직학적 또는 세포학적 진단을 받아야 합니다. 신경내분비종양, 선방세포 및 선편평세포암종 환자는 제외된다. 모든 질병은 기본 종양 평가 양식에 평가되고 문서화되어야 합니다.
  • 환자는 다음 돌연변이 중 하나를 가지고 있어야 합니다. 양성 및/또는 유해한 것으로 정의된 CLIA(Clinical Laboratory Improvement Act) 인증 실험실에서 테스트된 BRCA 1 또는 2의 생식계열 돌연변이(즉, 병원성 또는 병원성 가능성이 있는 변종). (참고: 종양 체세포 돌연변이가 있는 환자는 자격이 없습니다.)

  • 환자는 포르말린 고정 파라핀 포매(FFPE) 조직 및 전혈의 표본 보관에 참여할 기회를 제공받아야 합니다. 환자가 보관 조직을 제출할 수 없는 경우 환자가 NCCN(National Comprehensive Cancer Network) 지침에 따라 표준 치료 생검을 받아야 하는 경우 환자에게 해당 생검에서 새로운 종양 조직을 제출할 기회를 제공해야 합니다. 참가자의 동의 하에 SWOG(Southwest Oncology Group) 표본 추적 시스템을 통해 표본을 수집하고 제출해야 합니다.
  • 환자는 이 연구의 연구 특성에 대해 고지를 받아야 하며 기관 및 연방 지침에 따라 서명하고 정보에 입각한 동의를 제공해야 합니다. 의사 결정 능력이 손상된 참가자의 경우 법적 권한을 가진 대리인이 해당 연방, 지역 및 캐나다 노사 관계 위원회(CIRB) 규정에 따라 연구 참가자를 대신하여 서명하고 정보에 입각한 동의를 제공할 수 있습니다.
  • Oncology Patient Enrollment Network(OPEN) 등록 프로세스의 일부로 치료 기관의 신원이 제공되어 이 연구에 대한 현재(365일 이내) 기관 검토 위원회 승인 날짜가 시스템에 입력되었는지 확인합니다.

제외 기준:

  • 환자는 올라파립 또는 제품의 부형제에 알려진 과민증이 없어야 합니다.
  • 환자는 올라파립 치료를 받는 동안 강하거나 중등도의 CYP3A 억제제 또는 유도제를 투여받을 계획이 없어야 합니다. 강력하거나 중등도의 CYP3A 억제제를 투여받는 환자는 올라파립을 투여받기 최소 2주 전에 사용을 중단해야 합니다. 강력하거나 중등도의 CYP3A 유도제를 투여받는 환자는 올라파립을 투여받기 최소 5주 전에 사용을 중단해야 합니다. 피해야 할 약물 목록에 대해 자주 업데이트되는 의료 참고 자료를 사용하여 약물을 확인해야 합니다.
  • 환자는 무작위 배정 전 42일 이내에 생백신을 접종받은 적이 없어야 하며 연구 치료를 받는 동안 및 30일의 후속 조치 기간 동안 생 바이러스 또는 생 세균 백신을 접종할 계획이 없어야 합니다. 생백신의 예에는 홍역, 볼거리, 풍진, 수두, 대상포진, 황열병, 광견병, Bacillus Calmette-Guerin(BCG) 및 장티푸스(경구) 백신이 포함되나 이에 국한되지 않습니다. 주사용 계절 인플루엔자 백신은 일반적으로 사멸 바이러스 백신이며 허용됩니다. 그러나 비강내 인플루엔자 백신(예: Flu-Mist)은 약독화 생백신이며 허용되지 않습니다. 코로나바이러스 질병 2019(COVID-19) 메신저 리보핵산(mRNA) 백신은 허용됩니다

환자는 각 주기의 1~21일에 올라파립 PO BID를 받고, 각 주기의 1일에 30분에 걸쳐 펨브롤리주맙을 IV 투여받습니다. 질병 진행이나 허용할 수 없는 독성이 없는 경우 최대 18주기 동안 21일마다 치료가 반복됩니다. 19주기부터 환자는 1~42일에 올라파립 PO BID를 투여받고, 각 주기의 1일차에 30분에 걸쳐 펨브롤리주맙 IV를 투여받습니다. 질병 진행이나 허용할 수 없는 독성이 없으면 주기는 42일마다 반복됩니다. 환자는 연구 기간 동안 CT 스캔 또는 MRI, 종양 생검 및 혈액 샘플 수집을 거칩니다.
