Denne randomiserte fase III-studien studerer testosteron for å se hvor godt det fungerer sammenlignet med placebo ved behandling av postmenopausale pasienter med artralgi (leddsmerter) forårsaket av anastrozol eller letrozol. Testosteron kan bidra til å lindre moderat eller alvorlig artralgi forbundet med bruk av aromatasehemmere, for eksempel anastrozol eller letrozol.

Dette er en randomisert, placebokontrollert fase III-studie som evaluerer subkutan testosteron for lindring av aromatasehemmerindusert artralgi. En parallell gruppedesign vil bli brukt for denne to-armsstudien: subkutant testosteron vs. placebo. Pasientene er stratifisert etter baseline smertescore (5-6 vs. 7-10) og alder (< 50 vs. 50-60 vs. > 60). Hovedmålet er å finne ut om testosteron vil redusere AI-indusert artralgi og tilhørende leddsymptomer. Det sekundære målet er å undersøke om testosteron vil ha en akseptabel sikkerhets- og tolerabilitetsprofil, med særlig referanse til androgene bivirkninger inkludert akne, hirsutisme og alopecia.

Pasientene følges opp til seks måneder som definert i protokollen.

Alder ≥ 18 år
Får anastrozol (1 mg) eller letrozol (2,5 mg) oralt én gang daglig, i ≥ 21 dager før registrering og planlegger å fortsette det gjennom hele studiens varighet
Kroppsmasseindeks (BMI) mellom 18 og 35 kg/m^2
Kvinner som har gjennomgått total mastektomi eller brystbevarende operasjon for primær brystkreft +/-kjemoterapi, +/-strålebehandling.
Må ha BÅDE ER- og PR-reseptor-positive svulster og BEGGE må være ≥26 % positive. Alternativt, hvis ER og PR bestemmes av Allred-score, må poengsummen være 5 eller høyere
Kvinner som er postmenopausale ved kirurgi, strålebehandling eller tilstedeværelse av naturlig amenoré ≥ 12 måneder
≥ 5/10 artralgi (i hender, håndledd, knær eller hofter) mens de behandles med anastrozol eller letrozol som pasienten opplever å være forårsaket av deres aromatasehemmer som definert i protokollen. Merk: Pasienter kan, eller kanskje ikke, ta ikke-opioide analgetika
Evne til å fylle ut spørreskjema(er) selv eller med assistanse
ECOG Ytelsesstatus (PS) 0, 1 eller 2
Villig til å gi informert skriftlig samtykke
Villig til å returnere til en allianseregistrerende institusjon for oppfølging
Villig til å gi blodprøver for korrelative forskningsformål
Laboratorieverdier før registrering som definert i protokollen:
1. Kreatinin ≤1,5 x ULN
2. Hemoglobin > 11 g/dL
3. WBC > 3,0
4. Blodplateantall > 100 000
5. SGOT (AST) ≤ 1,5 x ULN

Ekskluderingskriterier:

Tilstedeværelse av gjenværende eller tilbakevendende kreft (lokalt eller metastatisk)
Diabetes mellitus eller glukoseintoleranse, definert som fastende glukose >125 mg/dL
Anamnese med koronarsykdom (angina eller hjerteinfarkt)
Pasienter på hormonbehandling (HRT) ≤ 4 uker før registrering. Dette inkluderer bruk av vaginal østrogenterapi.
Kjent overfølsomhet overfor en hvilken som helst komponent av testosteron.
Forlenget systemisk kortikosteroidbehandling, bortsett fra lokal bruk (f.eks. for utslett), inhalerte sprayer (f.eks. for obstruktive luftveissykdommer), øyedråper eller lokal insersjon (f.eks. intraartikulært). Merk: Kort varighet (< 2 uker) av systemiske kortikosteroider er tillatt (f.eks. for kronisk obstruktiv lungesykdom), men ikke innen 30 dager før registrering.

Anamnese med dyp venetrombose eller tromboemboli
