Randomisert fase II-studie av 2. linje FOLFIRI versus modifisert FOLFIRI med PARP-hemmer ABT-888 (Veliparib) (NSC-737664) i metastatisk bukspyttkjertelkreft

Denne randomiserte fase II-studien studerer hvor godt modifisert irinotekanhydroklorid, leukovorinkalsium, fluorouracil (FOLFIRI) og veliparib som en andre behandlingslinje fungerer sammenlignet med FOLFIRI ved behandling av pasienter med kreft i bukspyttkjertelen som har kommet tilbake etter en periode med bedring (metastatisk). PRIMÆRE MÅL:

I. SEKUNDÆRE MÅL:

I. For å evaluere hyppigheten og alvorlighetsgraden av toksisitet assosiert med hver av behandlingsarmene i denne pasientpopulasjonen.

II. For å evaluere progresjonsfri overlevelse (PFS) i hver av behandlingsarmene i denne pasientpopulasjonen.

OVERSIKT: Pasienter er randomisert til 1 av 2 behandlingsarmer.

ARM I: Pasienter får veliparib oralt (PO) to ganger daglig (BID) hver 12. time på dag 1-7, irinotekanhydroklorid intravenøst (IV) over 90-120 minutter på dag 3, leukovorin kalsium IV over 90-120 minutter på dag 3 , og fluorouracil IV over 46 timer på dag 3-5.

ARM II: Pasienter får irinotekanhydroklorid IV over 90-120 minutter på dag 1, leucovorin kalsium IV over 90-120 minutter på dag 1, og fluorouracil IV-bolus over 15 minutter på dag 1 og deretter over 46 timer på dag 1-3.

I begge armer gjentas kursene hver 14. dag i fravær av sykdomsprogresjon eller uakseptabel toksisitet.

Etter avsluttet studiebehandling følges pasientene opp hver 3. måned i 3 år.

Pasienter må ha histologisk eller cytologisk dokumentert pankreasadenokarsinom; pasienter med nevroendokrine svulster i bukspyttkjertelen, lymfom i bukspyttkjertelen eller ampullær kreft er ikke kvalifisert
Pasienter må ha metastatisk sykdom som er målbar; datatomografi (CT)-skanning eller magnetisk resonansavbildning (MRI) brukt for å vurdere målbar sykdom må ha blitt fullført innen 28 dager før registrering; CT-skanninger eller MR-er som brukes til å vurdere ikke-målbar sykdom, må være fullført innen 42 dager før registrering; all sykdom må vurderes og dokumenteres på Baseline Tumor Assessment Form

Tidligere systemisk terapi og kjemoradioterapi for behandling av resektabel, borderline-resekerbar eller lokalt avansert ikke-opererbar sykdom er tillatt og teller ikke mot tidligere behandling for metastatisk sykdom
Pasienter som fikk systemisk behandling med gemcitabin/nab-paclitaxel for resektabel eller borderline/lokalt avansert ikke-opererbar sykdom og progredierte med metastatisk sykdom innen 3 måneder etter siste dose av systemisk terapi er kvalifisert
Pasienter må ha fullført systemisk terapi minst 14 dager før registrering, enhver kirurgisk prosedyre må være utført minst 14 dager før registrering, og strålebehandling må være fullført minst 7 dager før registrering; Pasienter må ha kommet seg etter alvorlige bivirkninger av tidligere terapier eller prosedyrer etter den lokale stedsetterforskerens oppfatning før registrering
