Tämä vaiheen III tutkimus tutkii, kuinka hyvin bupropioni vähentää syöpään liittyvää väsymystä vaiheen I-III rintasyövästä selviytyneillä. Bupropioni on lääke, jota käytetään masennuksen hoitoon sekä tupakoinnin lopettamiseen. Syöpä ja sen hoito voivat aiheuttaa väsymystä. ENSISIJAISET TAVOITTEET:

I. Määrittää kontrolloidusti vapautuvan bupropionihydrokloridin (bupropionin) tehokkuus lumelääkkeeseen verrattuna väsymyksen vähentämisessä kaksoissokkoutetussa, lumekontrolloidussa, satunnaistetussa kliinisessä tutkimuksessa rintasyövästä eloonjääneillä, joilla on väsymystä.

TOISIJAISET TAVOITTEET:

I. Arvioida bupropionin tehoa lumelääkkeeseen verrattuna masennukseen ja elämänlaatuun väsyneillä rintasyövästä selviytyneillä.

II. Arvioida bupropionin siedettävyyttä rintasyövästä selviytyneillä väsyneillä.

TUTKIMUSTAVOITTEET:

I. Arvioida bupropionin tehoa lumelääkkeeseen verrattuna oireisiin ja kognitioon väsyneillä rintasyövästä selviytyneillä.

II. Tutkia bupropionin vaikutuksia oletettuihin syöpään liittyvän väsymyksen mekanismeihin.

III. Tutkia CYP2B6-genotyypin yhteyksiä bupropionin aineenvaihduntaan ja väsymyksen muutoksiin.

YHTEENVETO: Potilaat satunnaistetaan yhteen kahdesta haarasta.

ARM I: Potilaat saavat kontrolloidusti vapautuvaa bupropionihydrokloridia suun kautta (PO) kerran päivässä (QD) enintään 13 viikon ajan ilman hyväksyttäviä toksisuutta.

VAARA II: Potilaat saavat lumelääkettä PO QD enintään 13 viikon ajan ilman hyväksyttävää toksisuutta.

Sisällyttämiskriteerit:

  • Sinulla on diagnosoitu vaiheen I-III rintasyöpä
  • Ilmoita viime viikon pahin väsymys kohtalaiseksi tai vaikeaksi (eli pistemäärä >= 4 asteikolla 0-10, seulontatoimenpiteet, kysymys 1)
  • olet suorittanut leikkauksen, sädehoidon ja/tai kemoterapian vähintään 2 kuukautta ennen ilmoittautumista (osallistujat voivat saada ylläpito-, kohdennettua tai hormonaalista hoitoa)
  • Pystyy lukemaan ja puhumaan englantia
  • Tällä hetkellä ei ole raskaana tai imetä. Poissulkemiskriteerit:

Mitattu funktionaalisen arvioinnin syöpähoidon kokonaispistemäärällä-yleinen (tosiasia-G) asteikko (sisältyy FACIT-F). Tosiasia-G koostuu neljästä ala-asteikosta: fyysinen hyvinvointi, toiminnallinen hyvinvointi), emotionaalinen hyvinvointi ja sosiaalinen hyvinvointi. Neljän osa -asteikon pisteet summataan tuottamaan kokonaispistemäärä 0–108 ja korkeammat pisteet osoittavat parempaa elämänlaatua.
