EINE EINARMIGE, PROSPEKTIVE, MULTIZENTRIERTE STUDIE ZUR BEWERTUNG DER SICHERHEIT UND WIRKSAMKEIT VON ISAVUCONAZOL ZUR PRIMÄRBEHANDLUNG CHINESISCHER PATIENTEN MIT INVASIVER PILZKRANKHEIT (IFD), DIE DURCH ASPERGILLUS-ARTEN ODER ANDERE FADENPILZE VERURSACHT WIRD
Diese Studie ist eine Commitment-Studie nach der Zulassung und soll die Sicherheit und Wirksamkeit von Isavuconazol in einer relativ größeren chinesischen Bevölkerung, die eine Isavuconazol-Behandlung nach der Markteinführung erhalten wird, weiter untersuchen.
Dies ist eine einarmige, prospektive, multizentrische Studie. Diese Studie sucht chinesische Patienten mit nachgewiesener, wahrscheinlicher oder möglicher invasiver Pilzerkrankung (IFD), die durch Aspergillus-Arten oder andere Fadenpilze verursacht wird. Alle Teilnehmer erhalten eine Behandlung mit Isavuconazol. Die längste Behandlungsdauer in dieser Studie beträgt 84 Tage (bis zu 180 Tage für Teilnehmer, bei denen IM diagnostiziert wurde).
Das primäre Ziel ist die Charakterisierung der Sicherheit und Verträglichkeit von Isavuconazol durch Beobachtung der bei der Behandlung auftretenden unerwünschten Ereignisse.

Einschlusskriterien:
- nachgewiesene, wahrscheinliche oder mögliche IFD, verursacht durch Aspergillus-Spezies, Mucorales-Spezies oder andere Fadenpilze
- Körpergewicht > 40 kg beim Screening
Ausschlusskriterien:
- entweder chronische Aspergillose, Aspergillom oder ABPA
- Fortgeschrittene HIV-Infektion mit CD4-Zahl < 200 oder Krankheit, die das erworbene Immunschwächesyndrom definiert
- Personen, die 5 Tage wahrscheinlich nicht überleben, oder Teilnehmer an mechanischer Beatmung
- schwere Leberfunktionsstörung (Child-Pugh-Klasse C)
- Familiäres Short-QT-Syndrom
- Gleichzeitige Anwendung von Efavirenz, Ritonavir, Etravirin, Rifampicin/Rifampin, Rifabutin, Nafcillin, Ketoconazol oder Johanniskraut in den 5 Tagen vor der ersten Verabreichung der Studienintervention

All-cause mortality included any death that occurred after first dose of study drug through Day 42 as following: a) known deaths: any death that occurred after first dose of study intervention through Day 42 and b) unknown deaths: unknown (not actual deaths but the numbers were used in calculating all-cause mortality rate): participant was censored and was included in the reported data for this outcome measure if participant's survival status was missing or the last known alive date was before Day 42.

Clinical laboratory abnormalities test criteria included, a) hematology: hemoglobin with primary criteria of <0.8* lower limit of normal (LLN), erythrocytes <0.8* LLN, platelets <0.5* LLN >1.75* upper limit of normal (ULN), leukocytes < 0.6* LLN and > 1.5* ULN, lymphocytes and neutrophils < 0.8* LLN and > 1.2* ULN.
b) Chemistry: bilirubin and direct bilirubin >1.5* ULN, aspartate aminotransferase, alanine aminotransferase and alkaline phosphatase >3.0* ULN, urea nitrogen, urea and creatinine >1.3* ULN, sodium <0.95* LLN and potassium<0.9*
