기본 목표:

I. 이전에 치료받지 않은 만성 골수성 백혈병(CML) 환자에서 치료 12개월 후 중단점 클러스터 영역-Abelson 쥐 백혈병 바이러스 암유전자 동족체 1(BCR-ABL) 전사체의 감소로 측정한 분자 반응률을 비교하기 위해 dasatinib 100mg/일 또는 imatinib(STI571, Gleevec)(imatinib mesylate) 400mg/일로 치료받은 만성기 환자.

II. 이마티닙(STI571, Gleevec®) 용량을 400mg/일에서 800mg/일로 늘리면 분자 반응 속도가 증가하는지 여부를 테스트하기 위해 12개월 치료 후 BCR-ABL 전사 감소로 측정한 바에 따르면, 만성 단계에서 이전에 치료되지 않은 CML.

III. 이마티닙 400mg/일, 이마티닙 800mg/일 및 다사티닙 100mg/일에 대한 세포유전학적 및 혈액학적 반응의 비율을 추정합니다.

IV. imatinib 및 dasatinib로 치료받은 CML 환자의 반응에 대한 유도체 (der)(9) 및 der(22) 염색체 결실의 예후 효과를 예비 방식으로 평가합니다.

개요: 환자는 3개의 치료군 중 1개로 무작위 배정됩니다.

  • 환자는 등록 전 28일 이내에 얻은 골수 흡인 및 생검 및 말초혈액수를 기준으로 만성기 CML이 있어야 합니다.
  • 환자는 필라델피아 염색체 또는 (9;22)의 변이체의 존재를 입증하는 말초 혈액 또는 골수의 세포유전학적 또는 분자 분석을 기반으로 CML로 처음 진단된 날짜로부터 180일 이내에 이 연구에 등록해야 합니다. 실시간 폴리머라제 연쇄 반응(RT-PCR)에 의해 Bcr-Abl에 대해 양성 전좌 또는 시험; 환자는 필라델피아 염색체 외에 이차 염색체 이상이 있을 수 있으며 적격 상태를 유지합니다.
