상세 설명
골격근은 체중의 약 40%를 차지하며 인체 전체 단백질의 약 50~75%를 함유하고 있습니다. 단백질의 회전율은 인체의 규칙적인 과정입니다. 건강한 성인의 경우 근육 단백질 합성과 근육 단백질 분해 사이의 상호 작용으로 인해 순 성장이나 근육량 손실이 발생하지 않습니다. 그러나 저울이 근육 단백질 분해 쪽으로 기울어지면 근육 소모가 발생할 수 있습니다. 이는 근육이 움직이고 자세를 유지하기 위해 화학적 에너지를 기계적 에너지로 변환하는 명백한 역할을 할 뿐만 아니라 다음과 같은 덜 분명한 역할에도 관여하기 때문에 부정적인 결과를 초래할 수 있습니다. 혈액 순환에서 잠재적으로 독성이 있는 물질을 제거합니다. 다른 조직을 위한 연료 생산; 뇌와 면역 체계에 연료를 공급하는 데 중요한 에너지와 질소 저장; 영양 실조, 기아, 부상 및 질병 중에 상처 치유를 촉진합니다. 따라서 근육은 신체적 독립뿐만 아니라 인체의 단순한 생존을 위해서도 중요하다. 실제로 체내 단백질이 30%만 손실되어도 호흡과 순환 장애가 발생하여 결국 사망에 이를 수 있습니다. 이 연구의 목적은 골격근 성장을 조절하는 생리적 및 세포적 기전을 조사하는 것입니다. 이 연구의 결과는 기능을 개선하고 질병 합병증을 줄이며 생존율을 높이는 방법으로 근육량을 유지하고 증가시키기 위한 향후 치료법을 개발하는 데 도움이 될 수 있습니다.
이 연구는 세 가지 개별 실험 중 하나 내에서 여러 치료 부문 중 하나에 무작위로 배정될 건강한 참가자를 등록합니다. 전반적으로, 세 가지 실험은 다음을 조사할 것입니다. 아미노산 보충제의 운동 및/또는 섭취; (2) 저강도 저항운동 시 혈압계 밴드로 혈류를 제한하면 궁극적으로 근육 단백질 합성이 일어나는지 여부; (3) 노화가 근육 단백질 합성과 관련된 감소된 생리적 및 세포적 기전과 관련이 있는지 여부와 그러한 감소가 운동 후 아미노산 보충제 섭취 또는 저강도 저항 운동 중 혈류 제한으로 극복될 수 있는지 여부.
어떤 치료군 참가자가 배정되었는지에 따라 아미노산 보충, 라파마이신 약물, 니트로프루시드 나트륨 및/또는 위약을 투여받을 수 있습니다. 그들은 또한 혈류 제한과 함께 고강도 저항 운동, 저강도 저항 운동 또는 저강도 저항 운동을 할 수 있습니다. 모든 참가자는 단일 8시간 연구 방문과 1주일 후 후속 방문에 참석합니다. 연구 방문 동안 참가자는 다음을 받게 됩니다: 활력 징후, 신장 및 체중 측정; 혈액 및 소변 샘플링; 이중 에너지 x-선 흡수측정법(DEXA) 스캔; 추가 혈액 샘플링, 근육 생검 및 할당된 개입을 포함하는 주입 연구. 후속 방문에는 주입 연구 중에 만들어진 모든 절개에 대한 평가가 포함됩니다.

제외 기준:
- 일상 생활 활동(ADL)의 손상, 연간 2회 이상의 낙상 이력 또는 지난 1년 동안 상당한 체중 감소로 결정되는 신체적 의존성 또는 허약함
- 중등도에서 고강도 유산소 운동 또는 저항 운동의 주 2회 이상의 세션으로 구성된 운동 훈련
- 심각한 심장, 간, 신장, 혈액 또는 호흡기 질환
- 말초 혈관 질환
- 당뇨병 또는 기타 치료되지 않은 내분비 질환
- 활성 암
