Yksityiskohtainen kuvaus
Käynti 1:
Tarkastelun ja suostumuksen allekirjoittamisen jälkeen kohteet seulotaan. Seulonta koostuu:
Täytä IIEF (International Index of Erectile Function) ja ruokatiheyskysely.
Kirjaa muistiin sairaushistoria, lääkkeet Mittaa pituus ja paino Vitals Ottaa verta kokonaistestosteronin, sukupuolihormoneja sitovan globuliinin ja TSH:n (kilpirauhasta stimuloiva hormoni) varalta.
Jos IIEF-pisteet ovat epänormaalit, koehenkilöitä kehotetaan ottamaan yhteyttä PCP:hen (primary care provider).
Koehenkilöihin otetaan yhteyttä laboratoriotestien tuloksista. Jos poikkeavaa, koehenkilöä kehotetaan ottamaan yhteyttä PCP:hen.
Jos tutkittavalla ei ole PCP:tä, PCP:n yhteystiedot annetaan. Koehenkilöt, jotka täyttävät sisällyttämis- ja poissulkemiskriteerit, satunnaistetaan joko sekvenssiin yksi tai sekvenssi kaksi. Ilmoittautuessaan koehenkilöt eivät muuta ruokailutottumuksia koko tutkimuksen ajan muutoin kuin kahden illan ja kahden täyden päivän aikana, jolloin ruokaa tarjotaan (katso alla).

Vierailu 2:
Koehenkilöt palaavat tutkimuskeskukseen aamulla paastottuaan (muu kuin juoneet vettä) edellisenä iltana kello 23 jälkeen. Koehenkilöt eivät harjaa hampaitaan tai käytä suuvettä ja/tai muita hengityksen raikasteita sinä aamuna. Antropomorfisia, verenpaine- ja sykemittauksia tehdään. Veri otetaan ja varastoidaan. Virtsa kerätään ja säilytetään. Aiheisiin opetetaan Rigiscan™-laitteen käyttöä. Koehenkilöt saavat yhden kasvipohjaisen illallisen. Koehenkilöt poistuvat sitten tutkimuskeskuksesta ja syövät normaalia ruokavaliota klo 17 asti. Loppuillasta klo 23 asti tutkittavat syövät vain tarjotun illallisen. Vesi on ainoa sallittu juoma klo 17 jälkeen. Illallinen tulee syödä klo 20.30, EST (itäinen normaaliaika). Koehenkilöt lähettävät tekstiviestinä kuvia kaikesta, mitä on kulutettu (syö tai juo) kello 17 jälkeen PI:lle tai tutkimuksen edustajalle, jolla tutkimusryhmän jäsen on teksti- tai suullinen kontakti kohteen kanssa vähintään kerran sinä iltana. Koehenkilöt paastoavat muuta kuin vettä klo 23 jälkeen. Sinä iltana koehenkilöt tekevät Rigiscan™-arvioinnin yön yli kotona. Päivän aikana koehenkilön nukkumaanmenoaika ja herätysaika tulee tallentaa. Tutkittava suostuu olemaan sängyssä Rigiscan™:n kanssa (muuhun kuin hätätilanteisiin tai tarvittaviin vessaan-matkoihin) vähintään seitsemän tuntia.
Vierailu 3:
Koehenkilöt palaavat tutkimuskeskukseen Rigiscan™-arvioinnin jälkeisenä aamuna ja tuovat Rigiscan™-laitteen. Koehenkilöt ovat paastonneet muuta kuin juoneet vettä edellisenä iltana kello 23.00 lähtien. Koehenkilöt eivät harjaa hampaita tai käytä suuvettä ja/tai muita hengityksen raikasteita sinä aamuna. Antropomorfisia, verenpaine- ja sykemittauksia tehdään. Veri otetaan ja varastoidaan. Virtsa kerätään ja säilytetään. Koehenkilöt saavat sitten yhden täyden päivän kasviperäistä aterioita. Koehenkilöitä rohkaistaan syömään aamiaista tutkimuskeskuksessa sen jälkeen, kun ensimmäiset antropomorfiset mittaukset on tehty, veri on otettu ja virtsa on kerätty. Koehenkilöt lähettävät tekstiviestinä kuvia kaikesta, mitä sinä päivänä kulutetaan PI:lle ja/tai tutkimuksen edustajalle, ja tutkimusryhmän jäsenellä on teksti- tai suullinen yhteys tutkittavaan vähintään kahdesti päivän aikana. Illallinen tulee syödä klo 20.30, EST. Koehenkilöt paastoavat muuta kuin vettä klo 23 jälkeen. Sinä iltana koehenkilöt tekevät Rigiscan™-arvioinnin yön yli kotona. Tutkittava suostuu olemaan sängyssä Rigiscan™:n kanssa (muuhun kuin hätätilanteisiin tai tarvittaviin vessaan-matkoihin) vähintään seitsemän tuntia.
Vierailu 4:
Koehenkilöt palaavat tutkimuskeskukseen Rigiscan™-arvioinnin jälkeisenä aamuna ja tuovat Rigiscan™-laitteen. Koehenkilöt eivät harjaa hampaita tai käytä suuvettä ja/tai muita hengityksen raikasteita sinä aamuna. Antropomorfisia, verenpaine- ja sykemittauksia tehdään. Veri otetaan ja varastoidaan. Virtsa kerätään ja säilytetään. Sitten on 8-12 päivän pesujakso, ja koehenkilöitä neuvotaan palaamaan tavallisiin ruokailutottumuksiin tämän huuhtoutumisjakson aikana.
Vierailu 5:
Pesujakson jälkeen koehenkilöt palaavat tutkimuskeskukseen aamulla paastottuaan (muu kuin juoneet vettä) edellisenä iltana kello 23 jälkeen. Koehenkilöt eivät harjaa hampaita tai käytä suuvettä ja/tai muita hengityksen raikasteita sinä aamuna. Antropomorfisia, verenpaine- ja sykemittauksia tehdään. Veri otetaan ja varastoidaan. Virtsa kerätään ja säilytetään. Koehenkilöt saavat yhden eläinperäisen illallisen. Koehenkilöt poistuvat sitten tutkimuskeskuksesta ja syövät normaalia ruokavaliota klo 17 asti. Loppuillasta klo 23 asti tutkittavat syövät vain tarjotun illallisen. Vesi on ainoa juoma, joka juodaan klo 17 jälkeen. Illallinen tulee syödä klo 20.30, EST. Veri otetaan ja varastoidaan. Koehenkilöt saavat sitten yhden täyden päivän eläinperäisiä aterioita. Koehenkilöitä rohkaistaan syömään aamiaista tutkimuskeskuksessa sen jälkeen, kun ensimmäiset antropomorfiset mittaukset on tehty, sylki on saatu ja veri otettu. Koehenkilöt lähettävät tekstiviestinä kuvia kaikesta, mitä sinä päivänä kulutetaan PI:lle ja/tai tutkimuksen edustajalle, ja tutkimusryhmällä on teksti- tai suullinen kontakti tutkittavaan vähintään kahdesti päivän aikana. Sarja kaksi:
Sekvenssi kaksi on identtinen sekvenssin kanssa, paitsi että kasvipohjaisten aterioiden sijaan tarjotaan eläinperäisiä aterioita. Koehenkilöille, jotka on satunnaistettu saamaan eläinpohjaisia aterioita sekvenssissä yksi ja sekvenssi kaksi, ovat identtisiä paitsi, että kasviperäisiä aterioita tarjotaan eläinpohjaisten aterioiden sijaan.

- Tutkittava antaa kirjallisen tietoon perustuvan suostumuksen ja HIPAA-valtuutuksen englanniksi ennen tutkimustoimenpiteiden suorittamista;
- Ei lääkehoitoa (muita kuin prn-MDI:itä (mittariannosinhalaattoreita) ja ei muutoksia mahdollisiin lisäravinteisiin)
- IIEF-pistemäärä on suurempi tai yhtä suuri kuin 22
- Koehenkilö on harrastanut peniksen ja emättimen välistä seksiä 4 viikon sisällä ilmoittautumisesta. Käytetty IIEF-pistemäärä validoidaan tämän aikaikkunan perusteella.
- Mies 18-29v
- Asuu työmatkan päässä Montefiore Health Systemistä
- Tutkittavan merkittävä muu (tarvittaessa) suostuu tukemaan tutkittavaa tutkimuksen aikana
- Kohde suostuu valokuvaamaan ja jakamaan kaikki ruokavalion aikana syödyt/juomat tuotteet
- Kohde sitoutuu välttämään kaikkia laittomia huumeita, tulehduskipulääkkeitä ja alkoholia vähintään kaksi päivää ennen Rigiscan™-tallennusta sekä Rigiscan™-tallennuspäivinä ja -iltaisin
- Kohde sitoutuu pidättymään seksuaalisesta toiminnasta vähintään 36 tuntia ennen Rigiscan™-sijoitusta ja olemaan olematta seksuaalista toimintaa Rigiscan™-tallennuspäivinä ja -iltaisin.
- Kohde sitoutuu olemaan katsomatta, lukematta tai muuten kuluttamatta eroottista tai pornografista materiaalia vähintään 36 tuntiin ennen Rigiscan™-tallennuspäiviä ja -iltoja sekä Rigiscan™-tallennuspäivinä ja -öinä.
- Tutkittava suostuu tulemaan Montefioreen Rigiscan™-koulutukseen
- Kohde suostuu kuluttamaan/juomaan vain sallittuja ruokia/juomia.
- Tutkittava suostuu osallistumaan kaikkiin henkilökohtaisiin vierailuihin Montefioressa, aloittamaan paaston kello 23.00 kutakin käyntiä edeltävänä iltana ja käymään läpi kaikki verikokeet/sylki/muut testit.
- Tutkittava suostuu olemaan käyttämättä suu- tai suuhuuhteluvesiä tai tarkoituksellisesti sylkemättä koko tutkimuksen ajan. Koehenkilöitä pyydetään pidättäytymään suuveden käytöstä vähintään 2 päivää ennen ilmoittautumista. Tutkittava suostuu olemaan harjaamatta hampaitaan jokaisen opintokäynnin päivän aamuna. Hampaiden harjaus on muuten sallittua.
- BMI alle 30, BMI suurempi tai yhtä suuri kuin 18,5, paino yli 110 lbs.
- Tutkittava sitoutuu noudattamaan opintomenettelyjä ja vierailuja.
- Aiheharjoituksia vähintään 15 minuuttia vähintään kaksi kertaa viikossa. Tutkittava suostuu välttämään harjoittelua edellisenä päivänä ja Rigiscan™-mittauspäivänä
Poissulkemiskriteerit:
- Asiaankuuluva ruoka-aineallergia
- Kasvissyöjä tai vegaaninen ruokavaliomalli
- Syömishäiriön historia; todisteita ruokariippuvuudesta
- Hypertensio ja/tai aiempi verenpainetauti lääkehoidon kanssa tai ilman
- BMI suurempi tai yhtä suuri kuin 30, BMI alle 18,5 tai paino enintään 110 paunaa
- Tunnettu krooninen sairaus, joka ei ole tulehduksellinen tuki- ja liikuntaelinsairaus ja astma
- Aiempi munuaissairaus tai hyperkalemia
- Tutkittava on saanut tutkimuslääkkeen 30 päivän sisällä ennen suostumuksen allekirjoittamista
