Badanie mające na celu poznanie leku (PF-07321332 lub Nirmatrelvir/Ritonavir) u zdrowych kobiet karmiących piersią
OTWARTE BADANIE FARMAKOKINETYCZNE NIRMATRELVIRU/RYTONAWIRU U ZDROWYCH KOBIET W KARMIĄCYCH ŻYCIU
Głównym celem tego badania jest zmierzenie poziomu aktywnego składnika badanego leku (nirmatrelwiru), który jest wydzielany w mleku kobiecym podawanym zdrowym kobietom karmiącym piersią. Badany lek składa się z dwóch leków, nirmatrelwiru i rytonawiru. Poszukujemy uczestniczek płci żeńskiej:
- Aktywnie karmiące piersią (laktacja) co najmniej 12 tygodni po porodzie;
- Wiek od 18 do 55 lat i nie jest obecnie w ciąży;
- Mieć wskaźnik masy ciała (BMI): 17,5 kg/m2; i całkowitą masę ciała > 50 kg (110 funtów).
Uczestnicy będą przyjmować badany lek doustnie łącznie 3 razy w ciągu 2 dni (2 dawki poranne i 1 dawka wieczorna) w klinice badawczej. Będziemy okresowo pobierać mleko matki od dnia 2 do 4, aby zmierzyć w nim poziom nirmatrelwiru i rytonawiru. Po około 28-35 dniach od ostatniej dawki zostanie przeprowadzona rozmowa kontrolna dotycząca bezpieczeństwa, aby monitorować reakcje uczestników na badany lek.

Opis
Kryteria przyjęcia:
- Zdrowe kobiety w okresie laktacji, które aktywnie karmią piersią lub odciągają mleko, co najmniej 12 tygodni po porodzie i obecnie nie są w ciąży w wieku od 18 do 55 lat
- Wskaźnik masy ciała (BMI): 17,5 kg/m2; i całkowita masa ciała > 50 kg (110 funtów)
- Niemowlęta kobiet włączonych do badania muszą być w stanie skutecznie karmić się butelką lub inną odpowiednią do wieku alternatywną metodą karmienia przed rozpoczęciem badania i muszą być w stanie tolerować mleko modyfikowane dla niemowląt, jeśli matka nie ma zapasu mleka matki mleko wystarczające na czas trwania badania
- Uczestnicy muszą wyrazić gotowość do tymczasowego zaprzestania karmienia piersią swoich niemowląt na łącznie 4,5 dnia (108 godzin)
- Uczestnicy muszą być gotowi do regularnego odciągania pokarmu z piersi przez cały okres badania i odciągania pokarmu zgodnie z harmonogramem mającym na celu utrzymanie laktacji przez cały okres badania
Kryteria wyłączenia:
- Pozytywny wynik testu (RT-PCR) na zakażenie SARS-CoV-2 w czasie badania przesiewowego lub dnia -1
- Dowody lub historia istotnych klinicznie ustaleń
- Historia choroby przebiegającej z gorączką lub zapalenia sutka w ciągu 5 dni przed podaniem pierwszej dawki badanego leku
- Uczestnicy, którzy otrzymali szczepionkę przeciwko COVID-19 w ciągu 7 dni przed badaniem przesiewowym lub przyjęciem, lub którzy mają zostać zaszczepieni szczepionką przeciwko COVID-19 w dowolnym momencie w okresie zamknięcia badania
- Historia zakażenia wirusem HIV, wirusowym zapaleniem wątroby typu B lub wirusowym zapaleniem wątroby typu C; pozytywny wynik testu na obecność wirusa HIV, HBsAg lub HCVAb. Dozwolone jest szczepienie przeciwko wirusowemu zapaleniu wątroby typu B
- Nieprawidłowe parametry życiowe, takie jak ciśnienie krwi, 12-odprowadzeniowy elektrokardiogram
- Historia nadużywania alkoholu i/lub używania nielegalnych narkotyków, używanie tytoniu w ilości przekraczającej 5 papierosów dziennie lub 2 gryzienia dziennie
- Oddanie krwi w ciągu 60 dni
