EN FASE I, FLERE DOSISER, ÅBEN LABEL FARMAKOKINETISK UNDERSØGELSE AF NIRMATRELVIR/RITONAVIR HOS SUNDE AMMENDE KVINDER

Hovedformålet med denne undersøgelse er at måle niveauet af aktiv ingrediens i undersøgelsesmedicinen (nirmatrelvir), der udskilles i human modermælk, når det gives til raske ammende kvinder. Studiemedicinen består af to lægemidler, nirmatrelvir og ritonavir. Vi søger kvindelige deltagere, der er:

  • Aktivt amme (ammende) mindst 12 uger efter fødslen;
  • Alder mellem 18 og 55 år og ikke i øjeblikket gravid;
  • Har et Body Mass Index (BMI): 17,5 kg/m2; og en samlet kropsvægt >50 kg (110 lb).

Deltagerne vil tage undersøgelsesmedicinen gennem munden i alt 3 gange over 2 dage (2 morgendoser og 1 aftendosis) på studieklinikken. Vi vil med jævne mellemrum indsamle modermælk fra dag 2 til 4 for at måle niveauet af nirmatrelvir og ritonavir i det. Der vil blive foretaget en sikkerhedsopfølgning omkring 28-35 dage fra den sidste dosis for at overvåge eventuelle reaktioner, deltagerne måtte have på undersøgelsesmedicinen.

Inklusionskriterier:

  • Raske, ammende kvinder, der aktivt ammer eller udpumper modermælk, mindst 12 uger efter fødslen og ikke er gravide mellem 18 og 55 år.
  • Body Mass Index (BMI): 17,5 kg/m2; og en samlet kropsvægt >50 kg (110 lb)
  • Spædbørn til kvinder, der er tilmeldt undersøgelsen, skal være i stand til at spise med succes fra en flaske eller en anden alderssvarende alternativ ernæringsmetode før undersøgelsens start og skal kunne tåle modermælkserstatning, hvis moderen ikke har en forsyning af opbevaret bryst mælk tilstrækkelig til at dække undersøgelsens varighed
  • Deltagerne skal være villige til midlertidigt at stoppe med at amme deres spædbørn i i alt 4,5 dage (108 timer)
  • Deltagerne skal være villige til regelmæssigt at pumpe bryster gennem hele undersøgelsen og udpumpe mælk i henhold til en tidsplan, der er designet til at opretholde amning gennem hele undersøgelsesperioden

Ekskluderingskriterier:

  • Positivt testresultat (RT-PCR) for SARS-CoV-2-infektion på screeningstidspunktet eller dag -1
  • Evidens for eller historie med klinisk signifikante fund
  • Anamnese med febril sygdom eller mastitis inden for 5 dage før den første dosis af undersøgelsesmedicin
  • Deltagere, der har modtaget en COVID-19-vaccine inden for 7 dage før screening eller indlæggelse, eller som skal vaccineres med en COVID-19-vaccine på et hvilket som helst tidspunkt i undersøgelsesperioden
  • Anamnese med HIV-infektion, hepatitis B eller hepatitis C; positiv test for HIV, HBsAg eller HCVAb. Hepatitis B-vaccination er tilladt
