Uno studio per conoscere la medicina (PF-07321332 o Nirmatrelvir/Ritonavir) in donne sane che allattano
UNO STUDIO DI FARMACOCINETICA IN APERTO DI FARMACOCINETICA DI FARMATRELVIR/RITONAVIR IN DONNE SENE IN ALLATTAMENTO, A DOSE MULTIPLA
Lo scopo principale di questo studio è misurare il livello del principio attivo del medicinale in studio (nirmatrelvir) che viene secreto nel latte materno umano quando viene somministrato a donne sane che allattano. Il medicinale in studio è costituito da due medicinali, nirmatrelvir e ritonavir. Cerchiamo partecipanti donne che siano:
- Attivo allattamento al seno (allattamento) almeno 12 settimane dopo il parto;
- Età compresa tra 18 e 55 anni e non attualmente incinta;
- Avere un indice di massa corporea (BMI): 17,5 kg/m2; e un peso corporeo totale> 50 kg (110 libbre).
I partecipanti assumeranno il medicinale in studio per via orale per un totale di 3 volte nell'arco di 2 giorni (2 dosi mattutine e 1 dose serale) presso la clinica dello studio. Raccoglieremo periodicamente il latte materno dal giorno 2 al 4 per misurare il livello di nirmatrelvir e ritonavir in esso. Una chiamata di follow-up sulla sicurezza verrà condotta circa 28-35 giorni dall'ultima dose per monitorare eventuali reazioni che i partecipanti potrebbero avere al medicinale in studio.

Criterio di inclusione:
- Donne sane in allattamento che allattano attivamente o estraggono latte materno, almeno 12 settimane dopo il parto e non sono attualmente in stato di gravidanza tra i 18 e i 55 anni
- Indice di massa corporea (BMI): 17,5 kg/m2; e un peso corporeo totale> 50 kg (110 lb)
- I neonati delle donne arruolate nello studio devono essere in grado di nutrirsi con successo con il biberon o con un altro metodo di alimentazione alternativo adeguato all'età prima dell'inizio dello studio e devono essere in grado di tollerare il latte artificiale se la madre non dispone di una scorta di latte materno immagazzinato latte sufficiente a coprire la durata dello studio
- I partecipanti devono essere disposti a interrompere temporaneamente l'allattamento al seno dei loro bambini per un totale di 4,5 giorni (108 ore)
- I partecipanti devono essere disposti a pompare regolarmente il seno durante lo studio ed esprimere il latte secondo un programma progettato per mantenere l'allattamento per tutto il periodo di studio
Criteri di esclusione:
- Risultato positivo del test (RT-PCR) per l'infezione da SARS-CoV-2 al momento dello screening o al giorno -1
- Evidenza o anamnesi di risultati clinicamente significativi
- - Storia di malattia febbrile o mastite nei 5 giorni precedenti la prima dose del farmaco in studio
- - Partecipanti che hanno ricevuto un vaccino COVID-19 entro 7 giorni prima dello screening o del ricovero o che devono essere vaccinati con un vaccino COVID-19 in qualsiasi momento durante il periodo di confinamento dello studio
- Storia di infezione da HIV, epatite B o epatite C; test positivo per HIV, HBsAg o HCVAb. È consentita la vaccinazione contro l'epatite B
- Segni vitali anormali come pressione sanguigna, elettrocardiogramma a 12 derivazioni
- Storia di abuso di alcol e/o uso di droghe illecite, consumo di tabacco superiore a 5 sigarette/giorno o 2 masticazioni/giorno
- Donazione di sangue entro 60 giorni
