Farmacotoxicología de los metabolitos del tricloroetileno: efecto a corto plazo del DCA sobre el catabolismo de tirosina in vivo y la expresión de MAAI
Este proyecto se centra en la cinética, el metabolismo y la toxicología humana del catabolismo del dicloroacetato (DCA) y la tirosina.
La hipótesis es que el metabolismo de la tirosina será mayor en sujetos que albergan la variante KRT para GSTz1/MAAI para los que DCA muestra una Km alta.

Criterios de inclusión:
- Adultos programados para cirugía electiva por enfermedad hepática benigna.
- Electrocardiograma normal e historial.
- Laboratorios de referencia normales
Criterio de exclusión:
- El embarazo
- anemia severa, definida como un hematocrito < 30%.
- diabetes mellitus
- insuficiencia renal, definida como una creatinina sérica > 1,5 mg/dl o un aclaramiento de creatinina < 60 ml/min
- enzimas hepáticas elevadas
- enfermedad psiquiátrica que requiere medicación
- cirrosis biliar primaria o cualquier otra forma de cirrosis
- hepatitis viral o esteatohepatitis no viral
- enfermedad coronaria, definida como la necesidad de la administración diaria de medicamentos antianginosos o como insuficiencia cardíaca de clase III o IV de la New York Heart Association
- malignidad de cualquier tipo en cualquier localización anatómica

Experimental: Haplotipos y metabolismo DCA
Hombres y mujeres sanos con diferentes haplotipos recibirán una infusión de leucina y tirosina.
Al día siguiente comienzan un curso de 5 días de dicloroacetato (DCA) a una dosis de 2,5 mcg/kg/día.
El día 6 regresan y reciben otra infusión de leucina y tirosina.
Después de un período de lavado de 30 días, el sujeto regresa y nuevamente recibe una infusión de leucina y tirosina.
Luego, el día 2, comienzan con una dosis de DCA de 25 mg/kg durante 5 días y luego regresan para la infusión final de leucina y tirosina.

A los sujetos se les administra una infusión de los aminoácidos leucina y tirosina. Al día siguiente comienzan un curso de cinco días de dicloroacetato (DCA). Al cabo de cinco días reciben otra infusión de tirosina y leucina. La farmacocinética de DCA se calcula después de la segunda infusión. |
