Turvallisuus- ja immunogeenisuustutkimus VLA15:stä, moniarvoisesta rekombinantti OspA-pohjaisesta rokotteehdokkaista Lymen borrelioosia vastaan: satunnaistettu, kontrolloitu, tarkkailijasokea vaiheen 2 tutkimus terveellä lasten ja aikuisten tutkimuspopulaatiolla

VLA15-221 on vaiheen 2 tutkimus, joka suoritetaan kahdessa osassa: päätutkimusvaihe (osa A) ja tehostevaihe (osa B). Tutkimuksessa verrataan kahden erilaisen perusrokotusohjelman turvallisuutta ja immunogeenisyyttä kolmella (kuukausi 0-2-6) tai kahdella (kuukausi 0-6) rokotuksella. Tutkimukseen osallistuu 600 tervettä 5-65-vuotiasta henkilöä. Mukaan otetaan koehenkilöt, joilla on ollut Lymen borrelioosi (aiempi Borrelia-infektio) sekä henkilöt, jotka eivät ole saaneet borreliaa. Yksityiskohtainen kuvaus

VLA15-221 on satunnaistettu, tarkkailijasokko, lumekontrolloitu, monikeskustutkimusvaiheen 2 tutkimus, joka koostuu kahdesta osasta: päätutkimusvaihe (osa A) ja tehostevaihe (osa B). Osassa A 600 5–65-vuotiasta koehenkilöä otetaan 1:1:1 kolmeen ryhmään: Ryhmä 1 rokotetaan VLA15:llä kuukausina 0–2–6, ryhmä 2 rokotetaan VLA15:llä kuukausina 0–6 ja lumelääkkeellä kuukaudessa 2 ja ryhmässä 3 rokotetaan lumelääkettä kuukaudessa 0-2-6. Osassa B osa koehenkilöistä saa tehosteinjektion VLA15:llä tai lumelääkevalmisteella 18. kuukaudessa.

    • 18–65-vuotiaat: kirjallinen tietoinen suostumus ennen tutkimukseen liittyviä toimenpiteitä
    • 5–17-vuotiaille koehenkilöille: tutkittavan laillisen edustajan (laillisten edustajien) kirjallinen tietoinen suostumus paikallisten vaatimusten mukaisesti ja koehenkilön kirjallinen tietoinen suostumus ennen tutkimukseen liittyviä toimenpiteitä.

Poissulkemiskriteerit:

  • Potilaalla on Lymen borrelioosiin (LB) liittyvä krooninen sairaus, aktiivinen oireinen LB tai hän on saanut hoitoa LB:hen viimeisten 3 kuukauden aikana ennen päivää 1;
  • Kohde sai aikaisemman rokotuksen LB:tä vastaan;
  • Koehenkilöllä oli punkin purema 4 viikon sisällä ennen päivää 1;
  • Tutkittavalla on lääketieteellinen historia tai hänellä on tällä hetkellä kliinisesti merkittävä sairaus;
  • Kohdeella on lääketieteellinen historia tai hänellä on tällä hetkellä neuro-inflammatorinen tai autoimmuunisairaus;
  • Potilaalla on tunnettu trombosytopenia, verenvuotohäiriö tai hän on saanut antikoagulantteja 3 viikkoa ennen päivää 1;
  • Kohde on saanut aktiivisen tai passiivisen immunisoinnin 4 viikon sisällä ennen päivää 1;
  • Potilas on saanut mitä tahansa muuta rekisteröityä tai rekisteröimätöntä lääkettä toisessa kliinisessä tutkimuksessa neljän viikon sisällä ennen rokotusta päivänä 1;
