Critères d'inclusion clés :

  1. Diagnostic de la maladie et traitement antérieur :

    1. Cohorte 1 (la cohorte de traitement de première intention du cancer du sein) :

      • Participantes présentant un adénocarcinome du sein non résécable, localement avancé, récurrent ou métastatique confirmé histologiquement ou cytologiquement et candidates à la chimiothérapie. La maladie localement récurrente ne doit pas se prêter à une résection à visée curative.
      • Récepteur 2 du facteur de croissance épidermique humain (HER2) IHC 3+ ou hybridation in situ positive sur le tissu tumoral d'archives ou l'échantillon de biopsie frais.
      • N'ont pas reçu de traitement anticancéreux systémique antérieur pour une maladie localement avancée non résécable ou métastatique.
    2. Cohorte 2 (cohorte de traitement de première intention de l'adénocarcinome de la jonction gastrique/gastro-oesophagienne) :

      • Adénocarcinome de l'estomac ou de la jonction gastro-œsophagienne non résécable, localement avancé, récidivant ou métastatique confirmé histologiquement ou cytologiquement
      • HER2 IHC 3+ ou HER2 IHC 2+ avec hybridation in situ positive sur le tissu tumoral d'archive ou l'échantillon de biopsie frais.
      • N'ont pas reçu de traitement anticancéreux systémique antérieur pour une maladie non résécable ou métastatique localement avancée, y compris tout taux de filtration glomérulaire estimé (EGFR) approuvé ou expérimental ou des agents ou vaccins anti-HER2, une chimiothérapie cytotoxique ou des inhibiteurs de point de contrôle
  2. Au moins 1 lésion mesurable telle que définie par RECIST Version 1.1
  3. Statut de performance de l'Eastern Cooperative Oncology Group (ECOG) ≤ 1

  2. Antécédents d'inhibiteurs de la tyrosine kinase / HER approuvés ou en cours d'investigation dans n'importe quel contexte de traitement

    un. sauf trastuzumab avec ou sans pertuzumab utilisé en situation néoadjuvante ou adjuvante pour la Cohorte 1

  3. Maladie leptoméningée active, métastases cérébrales non traitées ou non contrôlées
  4. Toute tumeur maligne active ≤ 2 ans avant la première dose du médicament à l'étude, à l'exception du cancer spécifique à l'étude dans cet essai et de tout cancer localisé ayant fait l'objet d'un traitement curatif (par exemple, cancer de la peau basocellulaire ou épidermoïde réséqué, cancer superficiel de la vessie, carcinome situ du col de l'utérus ou du sein)
  5. Toute condition nécessitant un traitement systémique avec des corticostéroïdes (> 10 mg par jour de prednisone ou équivalent) ou un autre médicament immunosuppresseur ≤ 14 jours avant la première dose du médicament à l'étude

Remarque : les participants qui suivent actuellement ou ont déjà suivi l'un des régimes de stéroïdes suivants ne sont pas exclus :

  1. Stéroïde de remplacement surrénalien (dose ≤ 10 mg par jour de prednisone ou équivalent)
  2. Corticostéroïde topique, oculaire, intra-articulaire, intranasal ou inhalé avec une absorption systémique minimale
  3. Cure courte (≤ 7 jours) de corticostéroïdes prescrits à titre prophylactique (p. ex., pour une allergie aux produits de contraste) ou pour le traitement d'une affection non auto-immune (p. ex., réaction d'hypersensibilité de type retardé causée par un allergène de contact)

Expérimental: Cohort 1A: Breast Cancer: Zanidatamab 30 mg/kg + Docetaxel
Participants with HER2+ breast cancer received zanidatamab 30 mg/kg intravenously (IV) and docetaxel 75 mg/m^2 IV once every 3 weeks (Q3W) for an initial period of up to six cycles. After cycle 6, continuation of docetaxel treatment was at the discretion of the investigator. Zanidatamab was administered until disease progression, intolerable toxicity, or another criterion for treatment discontinuation was met.

Participants also received CAPOX chemotherapy (1000 mg/m^2 oral capecitabine twice daily for 14 days in each cycle and 130 mg/m^2 oxaliplatin IV once every 3 weeks) for up to 6 six cycles. After cycle 6, oxaliplatin was discontinued and continuation of capecitabine as maintenance treatment was at the investigator's discretion.

Participants also received CAPOX chemotherapy (1000 mg/m^2 oral capecitabine twice daily for 14 days in each cycle and 130 mg/m^2 oxaliplatin IV once every 3 weeks) for up to 6 six cycles. An AE is defined as any unfavorable and unintended sign (including an abnormal laboratory finding), symptom, or disease (new or exacerbated) temporally associated with the use of study treatments, whether considered related to study treatments or not.

A serious AE (SAE) was any untoward medical occurrence that, at any dose - resulted in death, - was life threatening, - required hospitalization or prolongation of existing hospitalization, - resulted in disability/incapacity, - was a congenital anomaly/birth defect.

Objective Response Rate (ORR)
