Criteri chiave di inclusione:
Diagnosi della malattia e trattamento precedente:
Coorte 1 (la coorte di trattamento del cancro al seno di prima linea):
- Partecipanti di sesso femminile con adenocarcinoma mammario non resecabile, localmente avanzato, ricorrente o metastatico confermato istologicamente o citologicamente e candidate alla chemioterapia. La malattia localmente ricorrente non deve essere suscettibile di resezione con intento curativo.
- Recettore 2 del fattore di crescita epidermico umano (HER2) IHC 3+ o ibridazione in situ positivo sul tessuto tumorale archiviato o sul campione bioptico fresco.
- - Non aver ricevuto una precedente terapia antitumorale sistemica per malattia localmente avanzata non resecabile o metastatica.
Coorte 2 (la coorte di trattamento di prima linea dell'adenocarcinoma della giunzione gastrica/gastroesofagea):
- Adenocarcinoma dello stomaco o della giunzione gastroesofagea non resecabile, localmente avanzato, ricorrente o metastatico confermato istologicamente o citologicamente
- HER2 IHC 3+ o HER2 IHC 2+ insieme all'ibridazione in situ positiva sul tessuto tumorale archiviato o sul campione bioptico fresco.
- Non hanno ricevuto una precedente terapia antitumorale sistemica per malattia localmente avanzata non resecabile o metastatica, inclusa qualsiasi velocità di filtrazione glomerulare (EGFR) stimata o approvata o agenti o vaccini anti-HER2, chemioterapia citotossica o inibitori del checkpoint
- Almeno 1 lesione misurabile come definita da RECIST Versione 1.1
- Performance Status dell'Eastern Cooperative Oncology Group (ECOG) ≤ 1
- Adeguata funzionalità degli organi come indicato dai seguenti valori di laboratorio durante lo screening:
- Frazione di eiezione ventricolare sinistra (LVEF) ≥ 50% al basale come determinato dall'ecocardiogramma o dalla scansione di acquisizione multigated (MUGA) (l'ecocardiogramma è il metodo preferito) entro 28 giorni prima della prima dose del farmaco in studio
Criteri chiave di esclusione:
- Terapia precedente con un anti-PD-1, anti-PD-L1, anti-PD-L2 o qualsiasi altro anticorpo o farmaco mirato specificamente alla costimolazione delle cellule T o ai percorsi del checkpoint
- Storia di inibitori della tirosina chinasi / HER approvati o sperimentali in qualsiasi contesto di trattamento
UN. eccetto trastuzumab con o senza pertuzumab utilizzato in ambito neoadiuvante o adiuvante per la Coorte 1
- Malattia leptomeningea attiva, metastasi cerebrali non trattate o non controllate
- Qualsiasi tumore maligno attivo ≤ 2 anni prima della prima dose del farmaco in studio, ad eccezione del tumore specifico oggetto di indagine in questo studio e di qualsiasi tumore localizzato che sia stato trattato in modo curativo (ad es. carcinoma cutaneo a cellule basali o squamose resecato, carcinoma superficiale della vescica, carcinoma in situ della cervice o del seno)
- Qualsiasi condizione che ha richiesto un trattamento sistemico con corticosteroidi (> 10 mg al giorno di prednisone o equivalente) o altri farmaci immunosoppressori ≤ 14 giorni prima della prima dose del farmaco in studio
Nota: i partecipanti che sono attualmente o sono stati in precedenza su uno dei seguenti regimi di steroidi non sono esclusi:
- Steroide sostitutivo surrenale (dose ≤ 10 mg al giorno di prednisone o equivalente)
- Corticosteroidi topici, oculari, intrarticolari, intranasali o inalatori con assorbimento sistemico minimo
- Breve ciclo (≤ 7 giorni) di corticosteroidi prescritti come profilassi (p. es., per allergia ai mezzi di contrasto) o per il trattamento di una condizione non autoimmune (p. es., reazione di ipersensibilità di tipo ritardato causata da allergeni da contatto)

Sperimentale: Cohort 1A: Breast Cancer: Zanidatamab 30 mg/kg + Docetaxel
Participants with HER2+ breast cancer received zanidatamab 30 mg/kg intravenously (IV) and docetaxel 75 mg/m^2 IV once every 3 weeks (Q3W) for an initial period of up to six cycles.
After cycle 6, continuation of docetaxel treatment was at the discretion of the investigator.
Zanidatamab was administered until disease progression, intolerable toxicity, or another criterion for treatment discontinuation was met.

Participants with HER2+ gastric / gastroesophageal junction cancer (GEJC) received zanidatamab 30 mg/kg IV and tislelizumab 200 mg IV once every 3 weeks until disease progression, intolerable toxicity, or another criterion for treatment discontinuation was met.
Participants also received CAPOX chemotherapy (1000 mg/m^2 oral capecitabine twice daily for 14 days in each cycle and 130 mg/m^2 oxaliplatin IV once every 3 weeks) for up to 6 six cycles.
After cycle 6, oxaliplatin was discontinued and continuation of capecitabine as maintenance treatment was at the investigator's discretion.

Participants also received CAPOX chemotherapy (1000 mg/m^2 oral capecitabine twice daily for 14 days in each cycle and 130 mg/m^2 oxaliplatin IV once every 3 weeks) for up to 6 six cycles.

An AE is defined as any unfavorable and unintended sign (including an abnormal laboratory finding), symptom, or disease (new or exacerbated) temporally associated with the use of study treatments, whether considered related to study treatments or not. A serious AE (SAE) was any untoward medical occurrence that, at any dose - resulted in death, - was life threatening, - required hospitalization or prolongation of existing hospitalization, - resulted in disability/incapacity, - was a congenital anomaly/birth defect. |

Objective Response Rate (ORR)

Complete Response (CR): Disappearance of all target lesions. Any pathological lymph nodes (whether target or non-target) must have reduction in short axis to < 10 mm. Partial Response (PR): At least a 30% decrease in the sum of diameters of target lesions, taking as reference the baseline sum diameters. |
