Kryteria przyjęcia:
- Uczestnik musi mieć ukończone 12 lat w momencie podpisania przez uczestnika badania lub opiekuna prawnego formularza świadomej zgody.
Tylko dla uczestników części eskalacyjnej:
- Potwierdzone rozpoznanie pierwotnej lub wtórnej AML [dowolny podtyp z wyjątkiem ostrej białaczki promielocytowej (APL)] zgodnie z klasyfikacją Światowej Organizacji Zdrowia (WHO). Pacjenci z AML muszą spełniać jedno z następujących kryteriów, a), b) lub c) i są ograniczeni do pacjentów bez dostępnej (lub niekwalifikujących się) terapii o znanej korzyści klinicznej.
a) Pierwotna niewydolność indukcji (PIF) AML, zdefiniowana jako choroba oporna na jeden z następujących czynników, i lub ii.
i) Intensywna próba wprowadzenia, na instytucję. Próby indukcji obejmują duże dawki i (lub) standardowe dawki cytarabiny ± antracykliny/antracenodion ± antymetabolit, z czynnikiem wzrostu lub bez czynnika wzrostu lub schematy zawierające terapię celowaną.
Przykłady obejmują między innymi:
- Jeden cykl schematu zawierającego wysoką dawkę cytarabiny (HiDAC).

- Nie kwalifikuje się do terapii indukcyjnej i ukończył ≥2 cykle któregokolwiek z następujących: lek hipometylujący (np. 5 azacytydyna lub decytabina) i/lub wenetoklaks, chemioterapia lub leki celowane.
Niekwalifikujący się do autologicznego przeszczepu komórek macierzystych (ASCT) i po ukończeniu ≥1 kursu terapii indukcyjnej.
- Potwierdzona diagnoza CD123 + B-ALL bez zmian pozaszpikowych, dla których nie ma dostępnej (lub nie kwalifikuje się) terapii ze znaną korzyścią kliniczną.
Tylko dla uczestników części rozszerzonej:
- Dla uczestników Kohorty A: Uczestnicy spełniający kryteria włączenia dla pacjentów z AML, którzy byli pierwotnie oporni (PIF) na wcześniejsze leczenie indukcyjne lub u których ER wystąpiło 6 miesięcy lub krócej po początkowej remisji podczas wcześniejszego leczenia indukcyjnego.
- Dla uczestników Kohorty B: Uczestnicy spełniający kryteria włączenia dla pacjentów z AML, u których wystąpił późny nawrót (LR), występujący ponad 6 miesięcy po początkowej remisji podczas wcześniejszego leczenia indukcyjnego.

Kryteria wyłączenia:
- Stan sprawności Eastern Cooperative Oncology Group (ECOG) >2 (≥18 lat). Skala Karnowskiego (16-17 lat)
- Historia aktywnego lub przewlekłego stanu autoimmunologicznego, który wymagał lub wymaga terapii.
- Drugi pierwotny nowotwór wymagający aktywnej terapii. Dozwolona jest uzupełniająca terapia hormonalna.
- Dowody na aktywną białaczkę ośrodkowego układu nerwowego w momencie rejestracji, potwierdzone badaniem cytologicznym lub patologicznym.
- Znany zespół nabytego niedoboru odporności (choroby związane z AIDS) lub choroba ludzkiego wirusa upośledzenia odporności (HIV) wymagająca leczenia przeciwretrowirusowego lub z czynnym zakażeniem wirusem zapalenia wątroby typu B lub C lub zakażeniem koronawirusem 2 (SARS-CoV-2) zespołu ostrej niewydolności oddechowej. Uczestnicy z historią zakażenia SARS-CoV-2 muszą zakończyć kliniczny powrót do zdrowia co najmniej 1 miesiąc przed rejestracją. - Wcześniejsze leczenie lekiem skierowanym przeciwko CD123.
- Wcześniejsze HSCT z nawrotem trwającym dłużej niż 3 miesiące można uwzględnić tylko w przypadku braku immunosupresji przez co najmniej 4 tygodnie i braku objawów choroby przeszczep przeciwko gospodarzowi (GVHD).
- Przyjmowanie w czasie pierwszego podania badanego produktu leczniczego (IMP) kortykosteroidu jako leku towarzyszącego z dawką kortykosteroidu >10 mg/dobę doustnego prednizonu lub ekwiwalentu,
- Wcześniejsze leczenie terapią komórkową, np. chimeryczną komórką T receptora antygenu (CAR-T) lub chimeryczną komórką NK receptora antygenu (CAR-NK).
- Jednoczesne leczenie innymi badanymi lekami.
- Podczas badania nie wolno stosować radioterapii, nawet jeśli ma ona charakter paliatywny.
- Profilaktyczne stosowanie krwiotwórczych czynników wzrostu (np. czynnik stymulujący tworzenie kolonii granulocytów (G-CSF), czynnik stymulujący tworzenie kolonii granulocytów i makrofagów (GM-CSF), erytropoetyna) podczas okresu obserwacji DLT wyłącznie w części dotyczącej zwiększania dawki. - Osoby umieszczone w instytucji ze względu na porządek regulacyjny lub prawny; więźniów lub uczestników, którzy są prawnie zinstytucjonalizowani.
- Kobiety w ciąży i karmiące piersią.
- Historia przeszczepów narządów miąższowych, w tym przeszczepu rogówki.
- Średni odstęp QTc (za pomocą obliczenia korekty Fridericia) >470 milisekund (ms) podczas badania przesiewowego.

For subgroup of participants with TD at baseline, the TI rate was defined as the percentage of participants who convert from baseline TD to TI during on treatment period; For subgroup of participants with TI at baseline, the TI rate was defined as the percentage of participants who remain TI during 56-day post baseline period during treatment.
