Eine klinische Studie zu Enlicitide Decanoat bei Menschen mit Leberfunktionsproblemen (MK-0616-030)
Eine offene klinische Einzeldosisstudie zur Bewertung der Pharmakokinetik von Enlicitide bei Teilnehmern mit Leberfunktionsstörung
Forscher haben ein neues Studienmedikament namens Enlicitide Decanoat als neue Möglichkeit entwickelt, die Menge an Low-Density-Lipoprotein-Cholesterin (LDL-C) im Blut einer Person zu senken. Enlicitide-Decanoat wird von nun an „Enlicitide“ genannt.
Der Zweck dieser Studie besteht darin, herauszufinden, was im Laufe der Zeit mit Enlicitid im Körper einer Person passiert (eine pharmakokinetische oder PK-Studie). Die Forscher werden vergleichen, was mit Enlicitid im Körper passiert, wenn es Menschen mit Leberfunktionsstörung (HI – das bedeutet, dass die Leber nicht richtig funktioniert) und Menschen mit guter Gesundheit verabreicht wird.
Diese Studie wird aus 2 Teilen bestehen. In Teil 1 wird Enlicitide an Menschen mit mittelschwerem HI und Menschen mit gutem Gesundheitszustand verabreicht. Nach Teil 1 entscheiden sich die Forscher möglicherweise dafür, Menschen mit leichter HI einzubeziehen und zu vergleichen, was mit Enlicitid im Körper passiert, mit Menschen, die sich in einem guten Gesundheitszustand befinden.

Einschlusskriterien:
Zu den wichtigsten Einschlusskriterien gehören unter anderem die folgenden:
Alle Teilnehmer:
- War vor Beginn der Studie mindestens 3 Monate lang Nichtraucher oder mäßiger Raucher (≤ 10 Zigaretten pro Tag oder gleichwertig).
- Hat einen Body-Mass-Index (BMI) ≥ 18,0 und ≤ 40,0 kg/m2
Teilnehmer mit mittelschwerer oder leichter HI:
- Diagnose einer chronischen (> 6 Monate) und stabilen (keine plötzlichen oder schweren Krankheitsepisoden aufgrund einer Verschlechterung der Leberfunktion in den letzten 2 Monaten) Leberinsuffizienz mit Anzeichen einer Leberzirrhose (Lebervernarbung) jeglicher Ursache.
- Ist im Allgemeinen bei guter Gesundheit, mit Ausnahme von HI.
Gesunde Kontrollteilnehmer:
- Medizinisch gesund ohne klinisch bedeutsame Krankengeschichte, körperliche Untersuchung oder klinische Laborprofile
Ausschlusskriterien:
Zu den wichtigsten Ausschlusskriterien gehören unter anderem die folgenden:
Alle Teilnehmer:
- Vorgeschichte einer Magen-Darm-Erkrankung, die die Nahrungs- und Arzneimittelaufnahme beeinträchtigen kann, oder ein Magenbypass oder eine ähnliche Operation.
- Geschichte von Krebs
- Konsumiert mehr als 3 Portionen alkoholische Getränke pro Tag.
- Befindet sich in einer Statin-Hintergrundtherapie.
Teilnehmer mit mittelschwerer oder leichter HI:
- Schwere Komplikationen einer Lebererkrankung innerhalb von 3 Monaten nach Studienbeginn.
