Een open-label, klinische studie met een enkele dosis om de farmacokinetiek van Enlicitide te evalueren bij deelnemers met een leverfunctiestoornis
Onderzoekers hebben een nieuw onderzoeksgeneesmiddel ontworpen, enlicitide decanoaat genaamd, als een nieuwe manier om de hoeveelheid lipoproteïnecholesterol met lage dichtheid (LDL-C) in iemands bloed te verlagen. Enlicitide-decanoaat wordt vanaf nu "enlicitide" genoemd,
Het doel van dit onderzoek is om te leren wat er in de loop van de tijd met enlicitide in het lichaam van een persoon gebeurt (een farmacokinetisch of PK-onderzoek). Onderzoekers zullen vergelijken wat er met enlicitide in het lichaam gebeurt als het wordt gegeven aan mensen met een leverfunctiestoornis (HI, wat betekent dat de lever niet goed werkt) en mensen die in goede gezondheid verkeren.
Deze studie zal uit 2 delen bestaan. In deel 1 wordt enlicitide gegeven aan mensen met matige HI en mensen die in goede gezondheid verkeren. Na deel 1 kunnen onderzoekers besluiten om mensen met milde HI op te nemen en te vergelijken wat er met enlicitide in het lichaam gebeurt met mensen die in goede gezondheid verkeren.

Inclusiecriteria:
De belangrijkste inclusiecriteria omvatten, maar zijn niet beperkt tot, het volgende:
Alle deelnemers:
- Is een niet-roker of matige roker (≤ 10 sigaretten per dag of gelijkwaardig) gedurende ten minste 3 maanden voorafgaand aan de start van het onderzoek
- Heeft een body mass index (BMI) ≥ 18,0 en ≤ 40,0 kg/m2
Deelnemers met matige of milde HI:
- Diagnose van chronische (> 6 maanden) en stabiele (geen plotselinge of ernstige ziekte-episodes als gevolg van verslechtering van de leverfunctie in de afgelopen 2 maanden) leverinsufficiëntie, met cirrose (leverlittekens) door welke oorzaak dan ook.
- Is over het algemeen in goede gezondheid, met uitzondering van HI.
Gezonde controledeelnemers:
- Medisch gezond zonder klinisch significante medische voorgeschiedenis, lichamelijk onderzoek of klinische laboratoriumprofielen
Uitsluitingscriteria:
De belangrijkste uitsluitingscriteria omvatten, maar zijn niet beperkt tot, het volgende:
Alle deelnemers:
- Voorgeschiedenis van gastro-intestinale aandoeningen die de absorptie van voedsel en geneesmiddelen kunnen beïnvloeden, of die een maagbypass of soortgelijke operatie heeft ondergaan.
- Geschiedenis van kanker
- Consumpeert meer dan 3 porties alcoholische dranken per dag.
- Krijgt statine-achtergrondtherapie.
Deelnemers met matige of milde HI:
- Ernstige complicaties van een leverziekte binnen 3 maanden na aanvang van het onderzoek.
