通常の老化は、脳血流と認知機能を低下させます。 老化はまた、認知機能に関連する脳内の機能的結合を変化させるようです。 観察研究は、定期的な身体活動が脳血流の増加と認知機能の改善に関連していることを示唆しています。 しかし、定期的な身体活動と加齢に伴う脳血流との機構的な関連性は不明です。 これに関連して、老化や運動トレーニングの状態が脳内の血流の神経血管結合を変化させるかどうかは不明です. したがって、この研究の全体的な目標は、重要な恒常性メカニズムであり、脳灌流の効果的な調節のマーカーである脳血管拡張能力の加齢に伴う変化を調べて、それが機械的に認知にどのように関連しているかを判断することです。 さらに、研究者は、人間の脳血管拡張と認知との関係に対する身体活動の潜在的な有益な効果を調査します。
研究の目的は次のとおりです。
- 脳血管拡張反応が健康な成人の年齢と運動トレーニングの状態によって影響を受けるかどうかを判断すること。
- 健康な成人の脳血管拡張反応に対するシクロオキシゲナーゼ阻害の効果を比較すること。
- 健康な成人におけるシクロオキシゲナーゼ阻害に対する神経血管逆調節応答を決定する。

包含基準:
- 18~35歳の若者
- 55~75歳の高齢者
- 非喫煙者
- 身体的にアクティブ(少なくとも 30 分間、週に 3 回以上の運動)
- 座りっぱなし (週に 1 時間以上の正式な運動なし)。
除外基準:
- 対象年齢外の方
- -肝臓、腎臓、または血液学的疾患の病歴または証拠がある;末梢血管疾患;脳卒中/神経血管疾患;糖尿病;高血圧
- 肝臓、腎臓、血液疾患を示す薬を服用してください。高血圧を含む心血管疾患;脳卒中/神経血管疾患;と糖尿病
- 体格指数 >34 kg/m2
- インドメタシンと相互作用する可能性のある医師が処方した薬を服用してください。 アンギオテンシン変換酵素阻害剤(ACE阻害剤)、アンギオテンシンII受容体遮断薬(ARB)、利尿薬); 2) 出血のリスクを高める薬 (例: ワルファリン、ヘパリン、クロピドグレル、リバロキサバン、その他の NSAIDs);または 3) 腎毒性の増加に関連する薬物 (例: シクロスポリン、タクロリムス)

実験的:薬
高炭酸ガス制御試験: 3 つの段階的な CO2 上昇が、室内空気とバランスを取りながら、毎回 2%、4%、および 6% の吸気 CO2 (FICO2) の分画濃度を追加することによって、患者に適用されます。 呼気終末 CO2 (PetCO2) が上昇し、各目標レベルで 3 分間一定に維持されます。 分時換気量 (VE) と PetCO2 の呼吸ごとの変化が測定されます。 薬物-インドメタシン: 健康なボランティアに、インドメタシン懸濁液を 1.2 mg/kg で経口投与します。 薬剤投与後、患者は 90 分間安静にします。 |
