Normale veroudering vermindert de cerebrale doorbloeding en cognitieve functie. Veroudering lijkt ook de functionele connectiviteit in de hersenen te veranderen, wat geassocieerd is met cognitief functioneren. Observationele studies suggereren dat regelmatige fysieke activiteit geassocieerd is met een hogere cerebrale doorbloeding en verbeterde cognitieve functie. De mechanistische verbanden tussen regelmatige lichaamsbeweging en cerebrale doorbloeding met het ouder worden zijn echter onbekend. In deze context is het onduidelijk of veroudering of trainingsstatus de neurovasculaire koppeling van de bloedstroom in de hersenen verandert. Het algemene doel van deze studie is dus om de leeftijdsgerelateerde veranderingen in cerebrale vasodilaterende capaciteit, een belangrijk homeostatisch mechanisme en marker voor effectieve regulatie van cerebrale perfusie, te onderzoeken om te bepalen hoe het mechanistisch is gekoppeld aan cognitie. Bovendien zullen de onderzoekers het potentiële gunstige effect van fysieke activiteit op de relaties tussen cerebrale vasodilatatie en cognitie bij mensen onderzoeken.
De onderzoeksdoelen zijn:
- Om te bepalen of cerebrale vaatverwijdende reacties worden beïnvloed door leeftijd en trainingsstatus bij gezonde volwassenen.
- Om het effect van cyclo-oxygenase-remming op cerebrale vasodilatator-responsen bij gezonde volwassenen te vergelijken.
- Om de neurovasculaire tegenregulerende respons op cyclo-oxygenase-remming bij gezonde volwassenen te bepalen.

Beschrijving
Inclusiecriteria:
- Jonge proefpersonen tussen 18-35 jaar oud
- Oudere onderwerpen tussen 55-75 jaar oud
- Niet-rokers
- Lichamelijk actief (oefen meer dan 3 keer per week gedurende minstens 30 minuten)
- Sedentair (geen formele oefening meer dan 1 uur per week).
Uitsluitingscriteria:
- Buiten de opgegeven leeftijdscategorie
- Aanwezig met voorgeschiedenis of bewijs van lever-, nier- of hematologische ziekte; perifere vaatziekte; beroerte/neurovasculaire ziekte; suikerziekte; hypertensie
- Neem medicijnen die wijzen op lever-, nier-, hematologische aandoeningen; hart- en vaatziekten, waaronder hypertensie; beroerte/neurovasculaire ziekte; en suikerziekte
- Lichaamsmassa-index >34 kg/m2
- Neem door een arts voorgeschreven medicijnen die kunnen interageren met indomethacine: 1) cardiovasculaire geneesmiddelen (bijv. angiotensine-converting-enzymremmer (ACE-remmers), angiotensine II-receptorblokkers (ARB's), diuretica); 2) geneesmiddelen die het risico op bloedingen verhogen (bijv. warfarine, heparine, clopidogrel, rivaroxaban, andere NSAID's); of 3) geneesmiddelen geassocieerd met verhoogde niertoxiciteit (bijv. ciclosporine, tacrolimus)
- Kwetsbare bevolkingsgroepen (d.w.z. zwangere vrouwen, gevangenen, personen die niet in staat zijn om toestemming te geven, enz.)

Experimenteel: Geneesmiddel
Controle-hypercapnische proeven: drie stapsgewijze CO2-verhogingen worden op de patiënt toegepast door toevoeging van een fractionele concentratie van ingeademde CO2 (FICO2) van 2%, 4% en 6% elke keer, gebalanceerd met kamerlucht. De end tidal CO2 (PetCO2) wordt verhoogd en gedurende drie minuten constant gehouden op elk doelniveau. Ademhalingsveranderingen in minuutventilatie (VE) en PetCO2 worden gemeten. Geneesmiddel-indomethacine: bij gezonde vrijwilligers wordt indomethacine-suspensie oraal toegediend in een dosis van 1,2 mg/kg. Na toediening van het medicijn rust de patiënt 90 minuten rustig uit. |
