세포감소 치료 경험이 없는 본태혈소판증가증 환자에서 Bomedemstat(MK-3543)과 Hydroxyurea의 효능과 안전성을 평가하기 위한 제3상, 무작위, 이중 맹검, 능동 비교 대조 임상 연구
이 연구의 목적은 세포감소 치료를 받은 경험이 없는 본태혈소판증가증(ET) 참가자를 대상으로 수산화요소와 비교하여 보메뎀스타트의 효능과 안전성을 평가하는 것입니다.
주요 목적은 지속성 임상혈액학적 반응(DCHR)과 관련하여 보메뎀스타트와 수산화요소를 비교하는 것입니다.
일차 가설은 bomedemstat가 DCHR과 관련하여 hydroxyurea보다 우수하다는 것입니다.

- 골수증식성 신생물에 대한 세계보건기구 기준 및 세포감소 치료 적응증에 기초한 본태성 혈소판 혈증(ET) 진단
- 골수 섬유증 등급을 매기기 위한 유럽 합의 기준의 수정된 버전에 따라 골수 섬유증 점수가 0등급 또는 1등급입니다.
- ET에 대해 이전에 세포감소 치료를 받은 적이 없습니다.
- 인간 면역결핍 바이러스(HIV)에 감염된 참가자는 항레트로바이러스 치료로 HIV를 잘 통제했습니다.
- B형 간염 표면 항원(HBsAg) 양성인 참가자는 최소 4주 동안 B형 간염 바이러스(HBV) 항바이러스 치료를 받았고 HBV 바이러스 수치가 감지되지 않는 경우 자격이 있습니다.
- C형 간염 바이러스(HCV) 감염 병력이 있는 참가자는 HCV 바이러스 수치가 감지되지 않는 경우 자격이 있습니다.

The PROMIS F SF-7a is a 7-item participant-reported assessment that measures both the experience of fatigue and the interference of fatigue on daily activities over the past week.
Response options are on a 5-point Likert scale, ranging from 1 = never to 5 = always.
The total score is used in the analysis and is obtained by summing keyed scores of all items.
Scores can range from 7 to 35, with higher scores indicating greater fatigue.

The MFSAF v4.0 is a 7-item participant-reported myelofibrosis symptom assessment which asks respondents to report symptom severity at its worst for each of the 7 items on a 0 (Absent) to 10 (Worst Imaginable) numeric rating scale.
The change from baseline in MFSAF total score for all symptoms will be presented.

For participants who demonstrate DCHR, DODCHR is defined as the time from the first documented evidence of confirmed reduction of platelet and WBC count until confirmed increase of platelet and WBC counts to above acceptable threshold, locally assessed to be due to ET after latest confirmed hematologic response date or thrombotic or major hemorrhagic events or disease progression to MF, MDS or AML.
