Um estudo randomizado, simples-cego e controlado por placebo para investigar a segurança, a tolerabilidade e a farmacocinética de doses únicas crescentes de BI 3000202 administradas como comprimidos a indivíduos saudáveis do sexo masculino e um cruzamento randomizado, aberto, de dose única e bidirecional Comparação da biodisponibilidade relativa de BI 3000202 como comprimido com e sem alimentos em indivíduos saudáveis do sexo masculino
A parte de dose única crescente (SRD) do estudo investiga segurança, tolerabilidade e farmacocinética do BI 3000202.
A parte do efeito do alimento (FE) é conduzida para avaliar o efeito do alimento na biodisponibilidade relativa da formulação BI 3000202.

Critério de inclusão
- Indivíduos saudáveis do sexo masculino de acordo com a avaliação do investigador, com base em um histórico médico completo, incluindo exame físico, sinais vitais (pressão arterial (PA), frequência de pulso (PR)), eletrocardiograma de 12 derivações (ECG) e laboratório clínico testes sem anormalidades clinicamente significativas
- Idade de 18 a 45 anos (inclusive)
- Índice de massa corporal (IMC) de 18,5 a 29,9 kg/m^2 (inclusive)
- Consentimento informado por escrito assinado e datado de acordo com o ICH-GCP e a legislação local antes da admissão no estudo
Critério de exclusão
- Qualquer achado no exame médico (incluindo BP, PR ou ECG) desviando do normal e avaliado como clinicamente relevante pelo investigador
- Medição repetida da pressão arterial sistólica fora da faixa de 90 a 140 milímetros de mercúrio (mmHg), pressão arterial diastólica fora da faixa de 50 a 90 mmHg ou frequência de pulso fora da faixa de 50 a 90 batimentos por minuto (bpm)
- Qualquer valor laboratorial fora do intervalo de referência que o investigador considere ser de relevância clínica
- Qualquer evidência de uma doença concomitante avaliada como clinicamente relevante pelo investigador
- Distúrbios gastrointestinais, hepáticos, renais, respiratórios, cardiovasculares, metabólicos, imunológicos ou hormonais
- Colecistectomia ou outra cirurgia do trato gastrointestinal que possa interferir na farmacocinética da medicação em estudo (exceto apendicectomia ou reparo de hérnia simples)
- Doenças do sistema nervoso central (incluindo, entre outros, qualquer tipo de convulsão ou acidente vascular cerebral) e outros distúrbios neurológicos ou psiquiátricos relevantes
- História de hipotensão ortostática relevante, desmaios ou desmaios Outros critérios de exclusão se aplicam.

FE part: In treatment Period 1, participants received a low dose III film-coated tablet BI 3000202, orally administered with 240 mL of water after an overnight fast of at least 10 hours (= reference R). In treatment Period 2, participants received a low dose III film-coated tablet BI 3000202, orally administered with 240 mL of water 30 minutes after consuming a high-fat, high-calorie breakfast consumed after an overnight fast of at least 10 hours (=test T). There was a minimum 3-day washout period between the two treatments. |

Number of any treatment-emergent adverse event assessed as drug-related by the investigator. Percentages were calculated using the total number of participants per treatment as denominator. MedDRA version 26.1 was used for reporting. All adverse events occurring up to 48 hours (2 days) after drug administration were assigned to treatment. Medical judgment was used to determine whether there was a reasonable possibility of a causal relationship between the AE and the given trial treatment, considering all relevant factors, including pattern of reaction, temporal relationship, de-challenge or re-challenge, confounding factors such as concomitant medication, concomitant diseases and relevant history. |
