Detaillierte Beschreibung
Aktuelle Daten deuten darauf hin, dass Johanniskrautzubereitungen hepatische Cytochrom-P450-Enzyme und Transportproteine induzieren können. Dies kann zu Arzneimittelwechselwirkungen führen.
Das Studiendesign ist Standard für DDI-Studien und basiert auf den regulatorischen Leitlinien der Food and Drug Administration (FDA) und der European Medicines Agency (EMA).
Ein Cocktail-Ansatz, bei dem mehrere Cytochrom P450 (CYP)- oder P-Glykoprotein (P-gp)-spezifische Sondenmedikamente verabreicht werden, wird verwendet, um gleichzeitig die Aktivitäten dieser Enzyme und des Transporters P-gp zu bestimmen.

Beschreibung
Einschlusskriterien:
- schriftliche Einverständniserklärung
- Kaukasische männliche oder weibliche Probanden im Alter zwischen ≥ 18 und ≤ 55 Jahren
- Körperlich und geistig gesund
- BMI zwischen ≥19 und ≤29 kg/m2 und Körpergewicht ≤90 kg
- Nichtraucher
- Bei Frauen muss der Schwangerschaftstest beim Screening und bei der Aufnahme negativ sein
Ausschlusskriterien:
- Bekannte oder vermutete Überempfindlichkeit gegen Studienmedikamente
- Vorgeschichte von klinisch signifikanten Krankheiten
- Positiver Test auf Hepatitis B, Hepatitis C oder HIV-Screening
- Bekannte Lichtempfindlichkeit

The "number" as provided in the tables is the ratio (in %) of geometric LS means of AUC of day 17 dosing samples (probe drug cocktail plus Ze117) vs. Day 1 dosing samples (probe drug cocktail alone). A one-way ANOVA model (with terms for subject and treatment) was applied to analyze the effect of Ze117 on the respective probe drug. Log transformed (base e) parameters were used within the model. The estimated treatment difference between day 17 and day 1 was transformed back to original scale and presented as the ratio between treatments including the corresponding 90% confidence interval. Effect limits were predefined as follows: (1) Mild effect limits (50-200%), extended bioequivalence limits (70-143%), bioequivalence limits (80-125%). |
