Beschreibung
Einschlusskriterien:
Die Teilnehmer müssen mindestens eines der folgenden Merkmale aufweisen:
- Dokumentierter Nachweis einer VTE vor dieser Studie
- Mindestens 60 Jahre alt
- Ein Body-Mass-Index (BMI) größer oder gleich 30 Kilogramm pro Quadratmeter (kg/m²) oder
- Alter 50 bis unter 60 Jahre und BMI 25 bis unter 30 kg/m²
- Die Teilnehmer müssen eine unzureichende Reaktion oder Unverträglichkeit gegenüber mindestens 1 krankheitsmodifizierenden Antirheumatika (DMARD) (synthetisch oder biologisch) haben.
Ausschlusskriterien:
Der Teilnehmer darf zuvor keinen Januskinase (JAK)-Hemmer verwendet haben oder mehr als 1 früheren TNF-Hemmer erhalten haben, der:
- abgesetzt wegen IR (Mangel oder Verlust der Wirksamkeit) wegen RA, oder
- wegen Intoleranz (AE) abgesetzt, wenn es für irgendeine Indikation verwendet wird
- Die Teilnehmerinnen dürfen nicht schwanger sein oder stillen
- Die Teilnehmer dürfen nicht mehr als eine VTE gehabt haben
- Die Teilnehmer dürfen nicht an Krebs erkrankt sein
- Die Teilnehmer dürfen keinen aktiven Herpes zoster, keine schwere Infektion, aktive Tuberkulose oder eine andere schwere Krankheit haben
- Die Teilnehmer dürfen innerhalb von vier Wochen vor Studienbeginn keinen Lebendimpfstoff erhalten haben
- Die Teilnehmer dürfen innerhalb von vier Wochen vor Studienbeginn an keiner anderen klinischen Studie teilgenommen haben
- Die Teilnehmer dürfen im vergangenen Jahr keinen IV-Drogenkonsum, anderen illegalen Drogenmissbrauch oder chronischen Alkoholmissbrauch gehabt haben

Participants received 2 milligrams (mg) of baricitinib once daily until the planned number of adjudicated primary endpoint events occurred or until the data monitoring committee (DMC) recommended early termination.
Those with inadequate response at 1-year after start of treatment with baricitinib 2 mg could receive rescue treatment with baricitinib 4 mg.

Total Duration of Treatment Exposure for Combined Baricitinib Dose, Individual Baricitinib Dose and TNF Inhibitor

Duration of exposure was calculated as:
TNF inhibitor exposure = date of last TNFi (adalimumab or etanercept) - date of first TNFi dose (adalimumab or etanercept) + 1, regardless of treatment cycling from adalimumab to etanercept or vice versa. - Total baricitinib exposure (2/4 mg) = date of last baricitinib (2mg or 4mg) treatment - date of first baricitinib (2mg or 4mg) dose + 1, regardless of rescue from baricitinib 2mg to baricitinib 4mg. Patient-years is calculated as sum of duration of exposure in days for all patients in dosing regimen divided by 365.25. |
