Diese klinische Phase-2-Studie wird die Wirksamkeit von Nirogacestat bei Granulosazelltumoren der Eierstöcke (OvGCTs) untersuchen. Nirogacestat ist ein Gamma-Sekretase-Hemmer (GSI), von dem angenommen wird, dass er das Wachstum und die Aktivität von Ovarial-Granulosa-Tumoren verringert.

Detaillierte Beschreibung

Granulosazelltumoren der Eierstöcke (OvGCTs) machen 5-7 % aller Eierstockkrebserkrankungen aus (ca. 1,5 bis 2.000 neu diagnostizierte Patientinnen/Jahr in den Vereinigten Staaten) und sind der häufigste Subtyp von Keimstrangtumoren der Eierstöcke (70 %). Es wird erwartet, dass die Behandlung von Granulosazelltumoren mit Nirogacestat die Notch-induzierte Granulosazellproliferation hemmt.

Dies ist eine multizentrische, einarmige, offene Phase-2-Behandlungsstudie zur Bestimmung der Wirksamkeit, Sicherheit, Verträglichkeit und Pharmakokinetik von Nirogacestat bei erwachsenen Teilnehmern mit rezidiviertem/refraktärem OvGCT. Die Teilnehmer werden die Behandlung bis zum Fortschreiten der Krankheit gemäß den Response Evaluation Criteria in Solid Tumors (RECIST) v1.1 oder einer inakzeptablen Toxizität fortsetzen.

Wichtige Einschlusskriterien:

  • Hat vor der ersten Dosis der Studienbehandlung einen histologisch bestätigten rezidivierenden Granulosazelltumor des Eierstocks vom Erwachsenentyp
  • Haben Sie dokumentierte radiologische Beweise für einen Rückfall nach mindestens einer systemischen Therapie, die nicht für eine Operation oder Bestrahlung zugänglich ist, und haben Sie eine messbare Krankheit nach RECIST v1.1-Kriterien
  • Eine ausreichende Knochenmark-, Nieren- und Leberfunktion haben, wie durch Screening-Besuchslaborwerte definiert

Wichtige Ausschlusskriterien:
