Beschreibung
Einschlusskriterien:
- Histologisch bestätigte fortgeschrittene solide Tumoren (protokolldefinierte ausgewählte solide Tumoren) mit messbaren Läsionen gemäß Response Evaluation Criteria In Solid Tumors Version 1.1 (RECIST v1.1), die als nicht für eine Operation oder andere kurative Behandlungen oder Verfahren geeignet gelten.
- Die Krankheit muss während oder nach der Behandlung mit mindestens einer vorherigen systemischen Chemotherapie fortschreiten.
- Leistungsstatusbewertung der Eastern Cooperative Oncology Group von 0 oder 1.
- Lebenserwartung > 12 Wochen.
- Bereitschaft, eine Schwangerschaft zu vermeiden.
Ausschlusskriterien:
- Bekannte zusätzliche bösartige Erkrankung, die fortschreitet oder eine aktive Behandlung erfordert.
- Behandlungsbedürftige Metastasen im Zentralnervensystem (ZNS) und/oder leptomeningeale Erkrankung.
- Toxizität aus einer früheren Therapie, die sich nicht auf die im Protokoll definierten Grenzwerte erholt hat.
- Vorheriger Erhalt eines Anti-PD-1-, Anti-PD-L1- oder Anti-PD-L2-Mittels; Behandlung mit einem Immunmodulator (z. B. CTLA-4, GITR, LAG3, TIM3, OX40, ICOS, IL-2, 4-1BB, CAR-T-Zelle).
- Vorherige Therapie mit antiangiogenen niedermolekularen TKIs, die auf den VEGF-Signalweg abzielen
- Teilnahme an einer anderen interventionellen klinischen Studie während der Einnahme von INCB099280.
- Beeinträchtigte Herzfunktion oder klinisch signifikante Herzerkrankung.
- Anamnese oder Anzeichen einer interstitiellen Lungenerkrankung, einschließlich nichtinfektiöser Pneumonitis.
- Vorliegen von Magen-Darm-Erkrankungen, die die Arzneimittelaufnahme beeinträchtigen können.
- Jede Autoimmunerkrankung, die in den letzten 5 Jahren eine systemische Behandlung erforderte.
- Diagnose einer primären Immunschwäche oder chronische systemische Steroidtherapie mit einer Tagesdosis von mehr als 10 mg Prednison oder einem Äquivalent.
- Aktive Infektion, die eine systemische Therapie erfordert.
- Vorgeschichte von Organtransplantationen, einschließlich Stammzelltransplantation.
- Erhalt systemischer Antibiotika innerhalb von 28 Tagen nach der ersten Dosis der Studienbehandlung.
- Der Einsatz von Probiotika ist während des Screenings und während des gesamten Studienbehandlungszeitraums verboten.

Experimental: Teil 1: Dosiseskalation
Bis zu 6 Dosen INCB099280, die zweimal täglich (BID) in Kombination mit Axitinib BID verabreicht werden, werden ausgewertet, um die Dosis(en) für die weitere Bewertung in der Dosiserweiterungsphase der Studie zu ermitteln.

Experimental: Teil 2: Dosiserweiterung
Nach Abschluss von Teil 1 werden die Teilnehmer in eine von zwei krankheitsspezifischen Kohorten eingeschrieben: Kohorte 1: Erwachsene mit klarzelligem gynäkologischen Krebs mit mindestens 50 % klarzelliger Histologie, deren Krankheit auf oder nach mindestens einer vorherigen Krebslinie fortgeschritten ist systemische Chemotherapie und sind keine Kandidaten für eine kurative Operation oder (Chemo-)Bestrahlung.
Kohorte 2: Erwachsene mit seltenen histologischen Subtyp-Epithelkarzinomen des gynäkologischen Trakts, deren Krankheit während oder nach mindestens einer vorherigen systemischen Chemotherapielinie fortgeschritten ist und nicht für eine kurative Operation oder (Chemo-)Bestrahlung in Frage kommen.
Für jede Kohorte in der Teil-2-Erweiterung können eine oder zwei Dosen aus Teil 1 ausgewählt werden.
