A Phase II/III Clinical Study of SIM0811 in the Treatment of Acute Ischemic Stroke
A Phase II/III, Multicenter, Randomized, Double-Blind, Placebo-Controlled Clinical Trial to Evaluate the Safety and Efficacy of SIM0811 Injection in Patients With Acute Ischemic Stroke
The study will evaluate the efficacy and safety of SIM0811 in participants with acute ischemic stroke who are present within 4.5 to 24 hours since last known well. The main questions it aims to answer are:
- Does SIM0811 improve overall functional outcome, measured by the ability to perform daily activities independently without help 90 days after treatment?
- What medical problems do participants have when taking SIM0811? Researchers will compare different doses of SIM0811 to a placebo (a look-alike substance that contains no drug) to see if SIM0811 works to treat acute ischemic stroke.
Participants will:
- Receive SIM0811 or a placebo .
- Receive standard medical care for stroke. On the first day, all participants will also take an aspirin pill or an aspirin placebo to keep the treatment assignment a secret.
- Stay in the hospital for a few days to undergo medical tests, including CT or MRI scans and blood draws.
- Return to the clinic for follow-up visits for up to 90 days to check their recovery and overall health.

Descrizione
Inclusion Criteria:
- Age ≥ 18 years.
- Time from last known well: 4.5 to 24 hours.
- Acute ischemic stroke in a supratentorial territory, with corresponding clinical manifestations, and symptomatic high-grade stenosis or complete occlusion of the culprit vessel, involving the middle cerebral artery (MCA) M1, M2, or distal branches, the anterior cerebral artery (ACA), or the posterior cerebral artery (PCA).
- NIHSS score of 6-25 for the current stroke.
- Pre-stroke mRS ≤ 1.
- Female participants of childbearing potential and male participants whose partners are of childbearing potential must agree to use a highly effective contraceptive method from the time of informed consent signature until 6 months after the last dose of investigational product, and to refrain from donating sperm/ova.
- A written informed consent form approved by the Ethics Review Committee/Institutional Review Board (IRB), voluntarily signed by the patient or their legally authorized representative
Exclusion Criteria:
- Planned to undergo endovascular treatment or planned to receive thrombolytic drug therapy
- NIHSS consciousness score 1a > 2
- .Head, neck, or aortic dissection
- History of intracranial hemorrhage
