Mesothelioom is een vorm van kanker die ontstaat uit sereuze oppervlakken, meestal als reactie op eerdere blootstelling aan asbest. Bij meer dan 80% van de slachtoffers van mesothelioom kan een geschiedenis van blootstelling aan asbest worden opgewekt. Blootstelling aan asbest alleen is echter niet voldoende om de ontwikkeling van mesothelioom te veroorzaken. Bijna 27 miljoen mensen in de VS werden tussen 1940 en 1979 op het werk blootgesteld aan asbest, maar elk jaar worden slechts 3.000 nieuwe gevallen van mesothelioom vastgesteld. Daarom is de hypothese van de onderzoekers dat genetische variatie naast blootstelling aan asbest en gastheerfactoren bijdragen aan de ontwikkeling van mesothelioom. Op basis van de voorlopige studies van de onderzoekers wordt geschat dat er een populatie van meer dan 1.000 proefpersonen met mesothelioom nodig is om een geldige GWAS uit te voeren.
Daarom is een multicenterbenadering nodig om gegevens en DNA te verzamelen over voldoende aantallen mesothelioom om het genetische risico adequaat te kunnen evalueren. Het doel van dit voorstel is om een consortium van mesothelioomonderzoekers te ontwikkelen om fenotypische gegevens en DNA-monsters te delen en om genoombrede associatiescanning (GWAS) uit te voeren.

Oprichting van een consortium van onderzoekers (6 locaties) voor het verzamelen van bloed voor kiembaan-DNA en demografische informatie van 1000 mesothelioompatiënten.
Tijdsspanne: Deelnemers zullen bij één gelegenheid van 30-60 minuten worden gezien om bloed af te nemen en demografische informatie op te halen. Het zal tot 2 jaar duren om 1000 proefpersonen met mesothelioom van de verschillende sites in te schrijven.

Demografische variabelen die worden verzameld zijn onder meer; geboortedatum, geslacht, leeftijd bij eerste blootstelling aan asbest, type blootstelling (beroepsmatig of omstander), gezondheidsgeschiedenis van de familie, persoonlijke medische geschiedenis, rookgeschiedenis, leeftijd bij diagnose, latentie, tumorlocatie en celtype.

GWAS zal worden uitgevoerd op het DNA van de 1000 proefpersonen met mesothelioom en vergeleken met 1000 leeftijd en asbestblootstelling gematchte controles vrij van persoonlijke voorgeschiedenis en familiegeschiedenis van kanker.
Tijdsspanne: Het duurt tot 1 jaar, na het verzamelen van de 1000 mesothelioommonsters, om de GWAS uit te voeren en te analyseren.

Gezien het significante risico op kanker anders dan de index mesothelioom bij beide proefpersonen en hun 1e graads familielid (bijna 3 maal voor broers en zussen, ouders en de mesothelioom proefpersonen zelf en 7 maal voor hun kinderen), is het doel SNP's te identificeren die betrokken zijn bij mesothelioom en andere veel voorkomende kankergevoeligheid.
