Een gecontroleerde klinische proef om het resultaat van immunotherapie te onderzoeken met behulp van precisie-T-cellen die specifiek zijn voor meervoudig veel voorkomend tumor-geassocieerd antigeen bij de behandeling van patiënten die een radicale operatie voor hepatocellulair carcinoom ondergingen
Doelstellingen:
Het doel van deze studie is het evalueren van de veiligheid en werkzaamheid van radicale chirurgie in combinatie met dendritische celprecisie meervoudige antigeen T-cellen bij het verminderen van het recidief en de uitzaaiing van leverkanker.
methoden:
Deze studie ontwerpt een nieuwe therapie met behulp van dendritische celprecisie meerdere antigeen T-cellen. Er zullen 60 postoperatieve patiënten met hepatocellulair carcinoom worden ingeschreven. Ze zijn willekeurig verdeeld in postoperatieve routinetherapiegroep en dendritische celprecisie multipele antigeen T-cellen gecombineerd met postoperatieve routinetherapiegroep. Dendritische celprecisie meervoudige antigeen-T-celbehandelingen zullen elke 3 weken worden uitgevoerd met in totaal drie perioden. De post klinische indicatoren zijn progressievrije overleving en algehele overleving.

Beschrijving
Inclusiecriteria:
- Leeftijd 18~65 jaar oud, man of vrouw
- Ondertekende geïnformeerde toestemming
- Diagnose van hepatocellulair carcinoom (HCC), radicale chirurgie werd uitgevoerd
- Het recidief van HCC werd na de operatie niet gevonden en er werd geen andere immunotherapie uitgevoerd
- De Eastern Cooperative Oncology Group (ECOG)-score ≤2
- Child-Pugh-score van leverfunctie ≤ 9
- Routinebloed voldoet aan de eisen
Uitsluitingscriteria:
- Verwachte totale overleving < 3 maanden
- Radicale chirurgie werd niet uitgevoerd of er werd een recidief gevonden na de operatie
- Leverfunctie is Childs Pugh C
- Had andere immunotherapie gekregen
- Andere ernstige ziekten: het hart, de longen, de nieren, de spijsvertering, het zenuwstelsel, psychische stoornissen, immuunregulerende ziekten, stofwisselingsziekten, infectieziekten, enz.
- Niet in staat of niet bereid om geïnformeerde toestemming te geven, of niet voldoen aan de testvereisten

Op basis van postoperatieve routinebehandeling zullen patiënten 3 cycli van dendritische cel-precisie meervoudige antigeen-T-cellen (DC-PMAT)-cellen ondergaan.

Postoperatieve routinebehandeling volgens de lokale routines van het ziekenhuis; DC-celsuspensie (1 x 107 DC + fysiologische zoutoplossing + 0,25% humaan serumalbumine) 1 ml voor elke infusie, subcutane injectie voor elke infusie 3 cycli, elke cyclus ontving twee infusies op dag 19, 20; 40, 41; 61, 62. PMAT-celsuspensie (1-6×109 PMAT + fysiologische zoutoplossing + 0,25% humaan serumalbumine) 300 ml voor elke infusie, IV (in de ader) voor elke infusie 3 cycli, elke cyclus kreeg één infusie op dag 21, 42, 63.
