Inclusiecriteria:

  • Gediagnosticeerd met Systemische Lupus Erythematosus (SLE) ten minste 24 weken voor het screeningsbezoek
  • Maak kennis met de European Alliance of Associations for Rheumatology (EULAR)/American College of Rheumatology (ACR) 2019 classificatiecriteria voor SLE
  • Een van de volgende: positieve antinucleaire antilichamen (ANA) ≥ 1:80 bij screening OF positieve anti-dsDNA OF positieve anti-Smith (anti-Sm) zoals bepaald door het centrale laboratorium bij screening

  • Totale systemische lupus erythematosus Disease Activity Index-2K (SLEDAI-2K)-score ≥ 6 punten en klinische SLEDAI 2K-score ≥ 4 punten met gewrichtsaandoening en/of cutane vasculitis en/of huiduitslag

    • Lupushoofdpijn, alopecia, organisch hersensyndroom en slijmvlieszweren moeten worden geregistreerd op SLEDAI 2K, indien geïndiceerd, maar tellen niet mee voor de punten die vereist zijn voor screening bij binnenkomst

  • Ten minste één SLE-achtergrondbehandeling (immunosuppressivum en/of antimalariamiddel) is vereist gedurende ≥ 12 weken vóór het screeningsbezoek, moet gedurende ≥ 8 weken vóór het screeningsbezoek een stabiele dosis hebben en moet stabiel blijven tot randomisatie en gedurende de hele studiedeelname
  • Achtergrondtherapie met orale corticosteroïden (OCS; prednison of equivalent) is toegestaan, maar niet vereist. Voor deelnemers die OCS gebruiken, moet de dosis stabiel zijn gedurende ≥ 2 weken vóór het screeningsbezoek, mag deze niet hoger zijn dan 30 mg/dag bij de screening en moet deze stabiel blijven tot het bezoek in week 4. Deelnemers kunnen zowel een OCS gebruiken als een antimalariamiddel en/of een immunosuppressivum

Uitsluitingscriteria:

  • Diagnose van door geneesmiddelen geïnduceerde SLE in plaats van idiopathische SLE
  • Andere auto-immuunziekten (bv. multiple sclerose, psoriasis, inflammatoire darmziekte, enz.) zijn uitgesloten. Deelnemers met type I auto-immuundiabetes mellitus, auto-immuunziekte van de schildklier, coeliakie of secundair syndroom van Sjögren zijn niet uitgesloten - SLE-overlapsyndromen waaronder, maar niet beperkt tot, reumatoïde artritis, sclerodermie en gemengde bindweefselziekte zijn uitgesloten
  • Actieve of onstabiele neuropsychiatrische manifestaties van lupus, inclusief maar niet beperkt tot elke aandoening gedefinieerd door BILAG A-criteria
  • Actieve, ernstige klasse III en IV, lupus nefritis die behandeling met cytotoxische middelen of een hoge dosis CS vereist of kan vereisen
  • Geschiedenis van aangeboren of verworven immunodeficiëntie
  • Bekende actieve infectie, of een ernstige infectie-episode waarvoor ziekenhuisopname of behandeling met parenterale (intramusculaire of IV) antimicrobiële middelen (bijv. antibiotica, antivirale, antischimmel- of antiparasitaire middelen) binnen 30 dagen na randomisatie vereist is, of behandeling met orale antimicrobiële middelen binnen 2 weken van randomisatie -Momenteel op elke therapie voor chronische infectie (bijv. Pneumocystis, herpes zoster, cytomegalovirus, invasieve bacteriële of schimmelinfecties of atypische mycobacteriën)
  • Meer dan 1 immunosuppressivum gebruiken bij screening
  • Alleen in Japan: deelnemers met een positief resultaat van de β-D-glucan-assay
