Efeitos da pravastatina no colesterol, inflamação e cognição na esquizofrenia
Estudo de Fase 4 dos Efeitos da Pravastatina nos Níveis de Colesterol, Inflamação e Cognição na Esquizofrenia
Este estudo envolve pessoas com esquizofrenia ou transtorno esquizoafetivo, que estão atualmente tomando medicamentos antipsicóticos. Alguns medicamentos antipsicóticos podem causar um aumento nos níveis de colesterol, o que pode levar à inflamação no corpo. A inflamação representa um risco no desenvolvimento de doenças cardíacas, diabetes e problemas com a função cerebral. O objetivo deste estudo é verificar se a pravastatina pode:
- Baixar o colesterol
- diminuir a inflamação
- Melhorar a cognição em pacientes com esquizofrenia

Descrição
Critério de inclusão:
- Macho ou fêmea
- Idade 18-68 anos
- Diagnóstico de esquizofrenia, qualquer subtipo, transtorno esquizoafetivo, qualquer subtipo ou transtorno esquizofreniforme
- Conformidade bem estabelecida com medicamentos ambulatoriais, incluindo seus medicamentos antipsicóticos
Critério de exclusão:
- Incapacidade de fornecer consentimento informado
- Abuso atual de substâncias e álcool
- Doença médica significativa, incluindo insuficiência cardíaca congestiva, doença cardiovascular grave, doença renal (creatinina sérica > 1,5), insuficiência hepática grave ou doença hepática ativa, anemia (hemoglobina <11,0 gm/dL), história de traumatismo craniano grave e músculo não tratado doença.
- Psiquiatricamente instável
- Mulheres com potencial para engravidar que estão grávidas, amamentando ou que não desejam ou não podem usar uma forma eficaz de controle de natalidade durante todo o estudo
- Indivíduos tratados com medicamentos anti-inflamatórios (incluindo aspirina diária e ibuprofeno), diuréticos tiazídicos; agentes que induzem a perda de peso e erva de São João serão excluídos do estudo
- História atual de doença tireoidiana não tratada
- Tratamento atual com insulina
- Indivíduos em tratamento com medicamentos como: colchicina, antifúngicos azólicos (fluconazol, cetoconazol, itraconazol); antibióticos macrólidos (claritromicina, eritromicina); Inibidores da protease do HIV (ritonavir, indinavir, saquinavir, nelfinavir) que inibem a enzima hepática CYP 450 3A
- Hipersensibilidade conhecida à pravastatina ou a qualquer um de seus componentes

A Bateria Cognitiva Consensual de Pesquisa de Medição e Tratamento para Melhorar a Cognição na Esquizofrenia (MATRICS) mede o funcionamento cognitivo em 7 domínios: velocidade de processamento, atenção/vigilância, memória de trabalho (não verbal e verbal), aprendizagem verbal, aprendizagem visual, raciocínio e resolução de problemas e cognição social. A pontuação composta é calculada pelo programa de computador MATRICS, que pondera igualmente cada uma das 7 pontuações de domínio. A faixa de pontuações compostas é de 20 a 80. Pontuações mais altas indicam níveis mais altos de funcionamento cognitivo, enquanto pontuações mais baixas indicam níveis mais baixos de funcionamento cognitivo. |
