Effetti della pravastatina su colesterolo, infiammazione e cognizione nella schizofrenia
Studio di fase 4 sugli effetti della pravastatina sui livelli di colesterolo, infiammazione e cognizione nella schizofrenia
Questo studio coinvolge persone con schizofrenia o disturbo schizoaffettivo, che stanno attualmente assumendo farmaci antipsicotici. Alcuni farmaci antipsicotici possono causare un aumento dei livelli di colesterolo, che può portare a infiammazioni nel corpo. L'infiammazione rappresenta un rischio per lo sviluppo di malattie cardiache, diabete e problemi con la funzione cerebrale. Lo scopo di questo studio è verificare se la pravastatina può:
- Abbassare il colesterolo
- Diminuire l'infiammazione
- Migliorare la cognizione nei pazienti con schizofrenia

Descrizione
Criterio di inclusione:
- Maschio o femmina
- Età 18-68 anni
- Diagnosi di schizofrenia, qualsiasi sottotipo, disturbo schizoaffettivo, qualsiasi sottotipo o disturbo schizofreniforme
- Ben consolidata compliance con i farmaci ambulatoriali, compresi i loro farmaci antipsicotici
Criteri di esclusione:
- Impossibilità di fornire il consenso informato
- Sostanze attuali e abuso di alcol
- Malattia medica significativa, tra cui insufficienza cardiaca congestizia, grave malattia cardiovascolare, malattia renale (creatinina sierica > 1,5), grave compromissione epatica o malattia epatica attiva, anemia (emoglobina <11,0 gm/dL), anamnesi di grave trauma cranico e muscolo non trattato malattia.
- Psichiatricamente instabile
- Donne in età fertile che sono incinte, che allattano o che non vogliono o non sono in grado di utilizzare una forma efficace di controllo delle nascite durante l'intero studio
- Soggetti trattati con farmaci antinfiammatori (inclusi aspirina giornaliera e ibuprofene), diuretici tiazidici; agenti che inducono la perdita di peso e l'erba di San Giovanni saranno esclusi dallo studio
- Anamnesi attuale di malattia tiroidea non trattata
- Attuale trattamento con insulina
- Soggetti in trattamento con farmaci quali: colchicina, antimicotici azolici (fluconazolo, ketoconazolo, itraconazolo); antibiotici macrolidi (claritromicina, eritromicina); Inibitori della proteasi dell'HIV (ritonavir, indinavir, saquinavir, nelfinavir) che inibiscono l'enzima epatico CYP 450 3A
- Ipersensibilità nota alla pravastatina o ad uno qualsiasi dei suoi componenti

The Measurement and Treatment Research to Improve Cognition in Schizophrenia (MATRICS) Consensus Cognitive Battery misura il funzionamento cognitivo all'interno di 7 domini: velocità di elaborazione, attenzione/vigilanza, memoria di lavoro (non verbale e verbale), apprendimento verbale, apprendimento visivo, ragionamento e risoluzione dei problemi e cognizione sociale. Il punteggio composito è calcolato dal programma per computer MATRICS, che pondera equamente ciascuno dei 7 punteggi di dominio. La gamma di punteggi compositi è 20-80. Punteggi più alti indicano livelli più alti o funzionamento cognitivo, mentre punteggi più bassi indicano livelli più bassi di funzionamento cognitivo. |
