Aktiv MMP-8 i oral væske til påvisning af oral og oropharyngeal cancer
Målet med denne observationsundersøgelse er at evaluere, om den aktive matrix metalloproteinase-8 (aMMP-8) mundskylletest kan hjælpe med at opdage orale og orofaryngeale kræftformer. Forskere målte aMMP-8-niveauer i mundskylleprøver indsamlet fra 77 kræftdeltagere og 50 ikke-kræftdeltagere i kliniske omgivelser.

Målet med denne prospektive undersøgelse er at evaluere, om den aktive matrix metalloproteinase-8 (aMMP-8) mundskylletest kan påvise og differentiere 72 orale og orofaryngeale kræftformer fra 50 kontroldeltagere.
I prøvetagningsprocessen blev patienterne bedt om at forskylle med postevand i 30 s. Efter et minuts venten blev de bedt om at skylle deres mund 30 s med 5 ml destilleret vand i testsættet og spytte det ud i et sterilt målebæger.
Prøveopløsningen blev hældt i åbningen af testkassetten placeret på testsystemet (ORALyzer®) for kvantitative resultater. Efter opnåelse af de kvantitative resultater i 5-6 minutter, blev kassetten straks fjernet for at udføre den kvalitative evaluering (Periosafe®).
Ekspressionen af aMMP-8 blev sammenlignet med forskellige faktorer, herunder alder, køn, rygestatus, parodontitis, tumorstørrelse, nodale eller distale metastaser, cancerstadium, histologisk grad og tumorsted. Statistisk analyse blev udført for at evaluere disse sammenhænge.

- Biopsi-bekræftet oral eller orofaryngeal cancer identificeret ved følgende internationale klassifikation af sygdomme (ICD-10) koder: C00-C06, C08-C10, C13 og C32 ifølge American Joint Committee on Cancer (AJCC) 8. udgave
- Voksen
Ekskluderingskriterier:
- In situ kræft
- Tidligere kræftdiagnose inden for fem år forud for undersøgelsen
- Brug af tetracykliner på prøveudtagningstidspunktet
- Fulgte ikke producentens instruktioner før mundskylleprøven (dvs. børste tænder før prøvetagning)

aMMP-8-niveauer blev målt kvantitativt i realtid ved hjælp af rapid point-of-care (PoC) stolesæt (Periosafe®, Dentognostics GmbH, Solingen, Tyskland) og en kvantitativ læser (Oralyzer®, Dentognostics GmbH, Solingen, Tyskland) til at vurdere mundskylleprøver fra patienter med mund- og mundhulekræft sammenlignet med raske forsøgspersoner. Tærsklen for et positivt kvantitativt aMMP-8-resultat er sat til 20 ng/ml, med aflæsninger på ≥20 ng/ml betragtes som positive, og de <19,9 ng/ml anses for negativ. |
