Descripción
Criterios de inclusión:
- Dispuesto y capaz de dar su consentimiento informado.
- Edad 18 a 64 años en la Selección.
- Diagnosticado con AUD de gravedad que puede beneficiarse de un tratamiento farmacológico, es decir, AUD moderado a grave.
- Mínimo de 10 días de consumo excesivo de alcohol en los 28 días anteriores a la Selección.
- No han transcurrido más de 14 días desde el último HDD o la finalización de la desintoxicación.
- Puede abstenerse del consumo de cualquier sustancia alcohólica durante las 72 horas previas a la dosificación.
- Dispuesto a abstenerse de usar drogas recreativas desde la selección hasta el final del estudio
- Dispuesto a abstenerse de fumar durante su tiempo en la clínica el día de la dosificación.
- Dispuesto a abstenerse de consumir drogas psicodélicas desde la selección hasta el final del estudio.
- Vivir en un alojamiento estable/seguro en la comunidad.
- En posesión de un teléfono móvil personal y capaz de designar al menos a una persona localizadora (p. ej., un miembro de la familia, un amigo o un mentor de recuperación), con una dirección verificable y un número de teléfono para ayudar con la organización de citas de seguimiento.
Criterio de exclusión:
- Antecedentes personales o familiares de primer grado de esquizofrenia, trastorno bipolar, trastorno psicótico, trastorno delirante, trastorno paranoide o trastorno esquizoafectivo.
- Cualquier trastorno psiquiátrico mayor, a excepción de la ansiedad y/o depresión leves o moderadas.
- Un diagnóstico clínico de trastorno de estrés postraumático.
- Ideación o comportamiento suicida en la selección.
- Uso regular o dependencia de otras drogas además de la cafeína o la nicotina.
- Cualquier uso autoinformado de compuestos psicodélicos en los últimos 6 meses.
- Historial de convulsiones.
- Pacientes que muestren signos de abstinencia de alcohol el día de la dosificación.
- Positivo para alcohol el día de la dosificación.
- Prueba de drogas en orina positiva para drogas ilícitas o drogas de abuso.
- Cualquier obstrucción nasal, bloqueo o síntomas de congestión.
- Cualquier antecedente personal o familiar de hipertermia maligna.
- Pacientes con un trastorno cardiovascular no controlado que, a juicio del Investigador, pueda interferir en la interpretación de los resultados del estudio o constituir un riesgo para la salud del paciente si participa en el estudio.
- Diabetes no controlada o insulinodependiente.

Effects on the Mystical Experience Questionnaire (MEQ-30)

The MEQ-30 was performed to assess the extent of a participant's mystical experience after receiving BPL-003 on Day 0 (dosing day).
The questionnaire comprises a list of 30 phenomena, with subscales to measure mystical, positive mood, transcendence of space and time, and ineffability factors, measured on a 0 to 5 scale (0=none; not at all to 5=extreme).
A 'total MEQ-30 score' was calculated for each participant as the average of all their responses.
The total MEQ-30 score is presented.
The higher the score, the greater the mystical experience.
A total score of ≥3 was considered to be a significant mystical experience.

Effects on the Ego Dissolution Inventory (EDI)

The EDI was performed to assess the extent of a participant's dissolution of ego after receiving BPL-003 on Day 0 (dosing day).
The inventory comprises 8 statements.
Participants rated their agreement to each statement by marking on a visual analogue scale from 0 ("no, not more than usually") to 100 ("yes, entirely or completely").
Higher scores are indicative of more intense ego dissolution.
In practice, this means participants with higher scores could have a reduced sense of having a distinct, separate self; greater feelings of unity with the environment or universe; a loss of self-referential thoughts or identity; or altered boundaries between 'self' and 'other'.
A 'total EDI score' was calculated for each participant as the average of their individual-item scores.
The total EDI score is presented.

Participants were classed as experiencing an ego dissolution if they had a total score of >50.0 out of 100 on the EDI scale.

Description of the BPL-003 Subjective Experience Data, From a Qualitative Interview

After BPL-003 dosing, participants had the option to participate in a one-to-one guided interview with independent researchers trained in microphenomenology methods to describe their psychedelic experience.
Temporal aspects of drug effects (e.g., sensory, cognitive, metacognitive, and sense of self/other/connectedness) were elicited through open-ended interview questions.
