Een fase 3, gerandomiseerde, dubbelblinde, placebogecontroleerde, multicenter studie om de werkzaamheid en veiligheid van Octreotide subcutaan depot (CAM2029) te beoordelen bij patiënten met acromegalie
Het doel van deze studie is om de werkzaamheid en veiligheid van CAM2029 bij patiënten met acromegalie te beoordelen.
Patiënten zullen worden gerandomiseerd naar CAM2029 of placebo, eenmaal per maand subcutaan toegediend gedurende 6 maanden.

Deelname Criteria
Onderzoekers zoeken naar mensen die aan een bepaalde beschrijving voldoen, de zogenaamde geschiktheidscriteria. Enkele voorbeelden van deze criteria zijn iemands algemene gezondheidstoestand of eerdere behandelingen.

Inclusiecriteria:
- Mannelijke of vrouwelijke patiënten, ≥18 jaar bij screening
- In staat om schriftelijke geïnformeerde toestemming te geven om deel te nemen aan het onderzoek voordat enige onderzoekgerelateerde procedures worden uitgevoerd
- Diagnose van acromegalie door historisch bewijs van (aanhoudende of recidiverende) acromegalie
- Behandeling met een stabiele dosis octreotide LAR of lanreotide ATG gedurende ten minste 3 maanden als monotherapie voorafgaand aan de screening
- IGF-1-waarden ≤1xULN bij screening
- Adequate lever-, pancreas-, nier- en beenmergfuncties
- Normaal ECG
Uitsluitingscriteria:
- GH ≥2,5 μg/L bij screening (cyclus)
- Medische behandeling voor acromegalie hebben ondergaan met pasireotide (binnen 6 maanden voorafgaand aan screening), pegvisomant (binnen 3 maanden voorafgaand aan screening), dopamine-agonisten (binnen 3 maanden voorafgaand aan screening) of andere onderzoeksgeneesmiddelen (binnen 30 dagen of 5 halfwaardetijden voorafgaand aan de screening [wat het langst is]
- Patiënten die octreotide LAR of lanreotide ATG gewoonlijk minder vaak dan elke 4 weken gebruiken (bijv. elke 6 weken of 8 weken)
- Patiënten met compressie van het chiasma opticum dat een gezichtsvelddefect veroorzaakt voor wie chirurgische ingreep geïndiceerd is
- Patiënten die binnen 1 maand na screening om welke reden dan ook een grote operatie/chirurgische therapie hebben ondergaan
- Patiënten die binnen 6 maanden voorafgaand aan de screening een hypofyse-operatie hebben ondergaan
- Patiënten die eerder zijn bestraald met de hypofyse
- Patiënten met slecht gecontroleerde diabetes mellitus (hemoglobine A1c >8,0%)

CAM2029 (octreotide subcutaan depot) 20 mg/1,0
ml voor een dosis van 20 mg, subcutane injectie eenmaal per maand, behandeling van zes maanden.
Als downtitratie vereist is, is 10 mg/0,5 ml voor een dosis van 10 mg beschikbaar.
