Descrizione

Criterio di inclusione:

  • Pazienti di sesso maschile o femminile, ≥18 anni allo screening
  • In grado di fornire il consenso informato scritto a partecipare allo studio prima che venga eseguita qualsiasi procedura correlata allo studio
  • Diagnosi di acromegalia mediante evidenza storica di acromegalia (persistente o ricorrente).
  • Trattamento con una dose stabile di octreotide LAR o lanreotide ATG per almeno 3 mesi come monoterapia prima dello screening
  • Livelli di IGF-1 ≤1xULN allo screening
  • Adeguate funzioni epatiche, pancreatiche, renali e del midollo osseo
  • ECG normale

Criteri di esclusione:

  • GH ≥2,5 μg/L allo screening (ciclo)
  • Hanno ricevuto cure mediche per l'acromegalia con pasireotide (entro 6 mesi prima dello screening), pegvisomant (entro 3 mesi prima dello screening), agonisti della dopamina (entro 3 mesi prima dello screening) o altri agenti sperimentali (entro 30 giorni o 5 emivite prima dello screening [qualunque sia più lungo]
  • Pazienti che di solito assumono octreotide LAR o lanreotide ATG meno frequentemente di ogni 4 settimane (ad es. ogni 6 settimane o 8 settimane)
  • Pazienti con compressione del chiasma ottico che causa qualsiasi difetto del campo visivo per i quali è indicato l'intervento chirurgico
  • Pazienti sottoposti a chirurgia maggiore/terapia chirurgica per qualsiasi causa entro 1 mese dallo screening
  • Pazienti sottoposti a intervento chirurgico ipofisario entro 6 mesi prima dello screening
  • Pazienti che hanno ricevuto una precedente irradiazione ipofisaria
  • Pazienti con diabete mellito scarsamente controllato (emoglobina A1c >8,0%)

CAM2029 (deposito sottocutaneo di octreotide) 20 mg/1,0 ml per dose da 20 mg, iniezione sottocutanea una volta al mese, sei mesi di trattamento. Se è necessaria la titolazione verso il basso, sono disponibili 10 mg/0,5 ml per una dose da 10 mg.

The variable of interest was considered missing only if no IGF-1 value could be obtained from either the Week 22 or the Week 24 sample. A composite strategy was assumed for intercurrent events, and a participant was considered as a non-responder if they discontinued IMP, or had the dose reduced prior to Week 22 (regardless of IGF-1 values), and/or was switched to rescue medication. ULN was derived from the participant's sex and age at screening. In order to account for all participants in the intention-to-treat (ITT) analysis set, multiple imputation was applied. The mean proportion of responders was calculated as an average of responders across the imputed datasets. The resulting proportion was multiplied by 100 to present "Percentage of participants".
