Een onderzoek naar mRNA-1345, een mRNA-vaccin dat zich richt op het respiratoir syncytieel virus, bij kinderen van 2 tot

Een fase 2, gerandomiseerde, waarnemersblinde studie om de veiligheid, reactogeniciteit en immunogeniciteit te evalueren van mRNA-1345, een mRNA-vaccin dat zich richt op het respiratoir syncytieel virus, bij kinderen van 2 tot 2 jaar.
Het doel van het onderzoek is om de veiligheid, reactogeniciteit en immunogeniciteit van mRNA-1345 te evalueren bij kinderen van 2 tot <5 jaar oud en bij kinderen met een hoog risico op respiratoir syncytieel virus (RSV) van 5 tot <18 jaar oud. om de selectie van het dosisniveau voor de volgende ontwikkelingsfase (fase 3) te informeren.

Belangrijkste inclusiecriteria:
Cohort 1:
- 2 tot <5 jaar oud, op het moment dat de geïnformeerde toestemming wordt ondertekend.
- Gezond, of met stabiele chronische aandoeningen die het risico op RSV-ziekte verhogen, volgens het klinische oordeel van de onderzoeker.
Cohort 2:
- 5 tot <18 jaar oud, op het moment dat de geïnformeerde toestemming wordt ondertekend.
- Deelnemers met stabiele chronische aandoeningen verhogen het risico op RSV-ziekte.
- Vrouwelijke deelnemers die zwanger kunnen worden, mogen aan het onderzoek deelnemen als de deelnemer: 1) een negatieve urinezwangerschapstest heeft bij de screening en op de dag van injectie (dag 1); 2) gedurende 28 dagen voorafgaand aan Dag 1 adequate anticonceptie heeft toegepast of zich heeft onthouden van alle activiteiten die tot zwangerschap zouden kunnen leiden; 3) ermee heeft ingestemd adequate anticonceptie voort te zetten tot 90 dagen na de injectie; en 4) momenteel geen borstvoeding geeft.
Belangrijkste uitsluitingscriteria:
- Acuut ziek of koortsig (temperatuur ≥38,0°Celsius [100,4°Fahrenheit]) binnen 72 uur voorafgaand aan of tijdens het screeningsbezoek of dag 1.
- Geschiedenis van een diagnose of aandoening die, naar het oordeel van de onderzoeker, de beoordeling van het onderzoek kan beïnvloeden of de veiligheid van de deelnemers in gevaar kan brengen.
- Heeft ≤ 14 dagen vóór de injectie van het onderzoeksvaccin (dag 1) een goedgekeurd of goedgekeurd vaccin ontvangen of is van plan dit te ontvangen, of is van plan om binnen 14 dagen na de injectie van het onderzoeksvaccin een goedgekeurd of goedgekeurd vaccin te ontvangen.
- Ontvangst van eventuele eerdere systemische immunosuppressiva of immuunmodificerende medicijnen. Korte cursussen (<7 dagen) met orale corticosteroïden zijn toegestaan als deze minimaal 3 maanden vóór inschrijving zijn voltooid.
- Ontvangst van monoklonale antilichamen tegen RSV binnen 6 maanden voorafgaand aan deelname aan het onderzoek.
- Heeft deelgenomen aan een interventioneel klinisch onderzoek binnen 28 dagen (6 maanden voor een onderzoek waarin een product zonder vergunning in deze leeftijdsgroep wordt beoordeeld) voorafgaand aan de dag van inschrijving of is van plan dit te doen tijdens deelname aan dit onderzoek.
Opmerking: Er kunnen andere, in het protocol gedefinieerde inclusie- en exclusiecriteria van toepassing zijn.

Solicited ARs were collected in an electronic diary (eDiary).
Local ARs: injection site pain, erythema (redness), swelling/induration (hardness); and axillary (underarm) swelling or tenderness ipsilateral to the side of injection.
Systemic ARs: fever, headache, fatigue, myalgia, arthralgia, nausea/vomiting, and chills.
Note, not all solicited ARs were considered adverse events (AEs).
Investigator reviewed whether the solicited AR was also to be recorded as an AE.

RSV-LRTD: Cough or difficulty breathing (Based on Investigator's observation; difficulty breathing included signs of wheezing, stridor, tachypnoea, chest in-drawing or subcostal or intercostal retractions) and peripheral oxygen saturation (SpO2) <95%, or respiratory rate (RR) increased and confirmed RSV infection.
RSV Severe-LRTD: Meeting the definition of RSV-LRTD and SpO2 <93%, or lower chest wall in-drawing.
RSV Very Severe LRTD: Meeting the definition of RSV-LRTD and SpO2 <90%, or failure to respond/unconscious. RSV Hospitalization: Confirmed RSV and hospitalized for acute medical condition.
