O estudo será realizado em duas etapas:
Estágio 1: Desescalonamento de idade de adultos para crianças e bebês
- Os participantes adultos receberão iNTS-GMMA Dose C (alta) ou uma vacina de controle por via intramuscular no Dia 1 e no Dia 57.
- As crianças participantes receberão a Dose B (média) ou a Dose C (alta) da vacina candidata ou o controle no Dia 1 e no Dia 57.
- Os participantes infantis (9 meses de idade) receberão a Dose A (baixa), a Dose B (média) ou a Dose C (alta) da vacina candidata ou o controle no Dia 1, Dia 85 e Dia 169.
- Os participantes infantis (6 semanas de idade) receberão a Dose A (baixa), a Dose B (média) ou a Dose C (alta) da vacina candidata ou o controle no Dia 1, Dia 85 (fase de preparação) e Dia 232 (Fase de reforço).
Estágio 2: Determinação da dose em bebês de 6 semanas de idade
-Bebês (6 semanas de idade) receberão um dos três níveis de dose (Dose A [baixa], Dose B [média] ou Dose C [alta]) da vacina candidata ou do controle no Dia 1, Dia 85 ( Fase de preparação) e Dia 232 (fase de reforço).

Critério de inclusão:
Todos os participantes (adultos, crianças, bebês com 9 meses de idade e bebês com 6 semanas de idade) serão inscritos no centro clínico em Gana e deverão satisfazer TODOS os seguintes critérios no início do estudo:
- Participantes e/ou pais/representantes legalmente aceitáveis (LAR) dos participantes que, na opinião do investigador, podem e irão cumprir os requisitos do protocolo (por exemplo, preenchimento dos cartões diários, devolução para visitas de acompanhamento).
- Consentimento informado escrito ou testemunhado/impresso com polegar obtido do(s) participante(s)/pai(s)/LAR(es) do participante antes da realização de qualquer procedimento específico do estudo.
- Participantes saudáveis, conforme estabelecido pelo histórico médico, exame clínico e investigações laboratoriais.
- Participantes que atendem aos requisitos de triagem.
- Participantes negativos para vírus da imunodeficiência humana (HIV), hepatite B e hepatite C.
Os participantes adultos devem satisfazer TODOS os seguintes critérios no início do estudo:
- Um homem ou mulher entre 18 e 50 anos de idade no momento da primeira administração da intervenção do estudo.
- Participantes do sexo feminino sem potencial para engravidar podem ser inscritas no estudo. O potencial não reprodutivo é definido como pré-menarca, laqueadura ou oclusão tubária bilateral atual, histerectomia, ovariectomia bilateral ou pós-menopausa.
- Participantes do sexo feminino com potencial para engravidar podem ser inscritas no estudo se a participante:
praticou contracepção adequada por 1 mês antes da administração da intervenção do estudo, e:
- tem um teste de gravidez negativo no dia da administração da intervenção do estudo, e
- concordou em continuar a contracepção adequada durante todo o período de tratamento e por 1 mês após a conclusão da série de administração de intervenção do estudo.
O cartão de Gana será usado como documento fonte para verificar as idades dos adultos.
As crianças participantes devem satisfazer TODOS os seguintes critérios no início do estudo:
- Um homem ou mulher entre 24 e 59 meses de idade no momento da primeira administração da intervenção do estudo.
- Vacinações infantis de rotina previamente concluídas, de acordo com o melhor conhecimento dos pais/LAR do participante.
- Nasceu após um período de gestação ≥37 semanas.
Os participantes infantis devem satisfazer TODOS os seguintes critérios no início do estudo:
- Um homem ou mulher com 6 semanas ou 9 meses de idade no momento da primeira administração da intervenção do estudo.
- Nasceu após um período de gestação ≥37 semanas.
- Nasceu de mãe soronegativa para HIV, vírus da hepatite B e vírus da hepatite C.
O Gráfico do Caminho para a Saúde será usado como documento de origem para confirmar as idades das crianças e bebés.
Critério de exclusão:
Condições médicas
- Exposição conhecida a S. Typhimurium ou S. Enteritidis durante o período que começa no nascimento para bebês e crianças, e aos 3 anos para adultos, conforme documentado nos registros do paciente
- História de qualquer reação ou hipersensibilidade que possa ser exacerbada por qualquer componente das intervenções do estudo.
- Hipersensibilidade, incluindo alergia, a medicamentos ou equipamentos médicos cuja utilização está prevista neste estudo.
- Condições clínicas progressivas, instáveis ou não controladas.
- Qualquer condição imunossupressora ou imunodeficiente confirmada ou suspeita, com base no histórico médico e exame físico
- Principais defeitos congênitos, conforme avaliado pelo investigador.
- Anormalidade funcional pulmonar, cardiovascular, hepática ou renal clinicamente significativa, aguda ou crônica, conforme determinado por exame físico ou testes laboratoriais de triagem.
- Doença aguda e/ou febre no momento da inscrição (febre é definida como temperatura ≥ 38,0°C).
- História recorrente ou distúrbios neurológicos não controlados ou convulsões.
- Qualquer anormalidade laboratorial hematológica e/ou bioquímica clinicamente significativa.
- Desnutrição definida como pontuação Z da OMS inferior a -2 DP.
- Infecção por malária definida como a presença de parasitas assexuados no sangue.
- Condições clínicas que representam contraindicação à vacinação intramuscular e coleta de sangue.
- Qualquer deficiência comportamental ou cognitiva ou doença psiquiátrica que, na opinião do investigador, possa interferir na capacidade do participante de participar do estudo.
- Qualquer outra condição clínica que, na opinião do investigador, possa representar risco adicional ao participante devido à participação no estudo.
Terapia prévia/concomitante
- História de recebimento de quaisquer vacinas experimentais iNTS ou GMMA na vida do participante.
- Uso de qualquer produto experimental ou não registrado que não seja as intervenções do estudo durante o período começando 30 dias antes da primeira dose das intervenções do estudo, ou seu uso planejado durante o período do estudo.
- Administração planejada/administração de uma vacina não prevista no protocolo do estudo no período que começa 14 dias antes de cada dose e termina 28 dias após a última dose da administração das intervenções do estudo, com exceção de vacinas contra gripe e vacinas administradas como parte de uma política de saúde pública campanha de vacinação.
Uma vacina não prevista no protocolo do estudo administrada durante o período que começa 14 dias antes da primeira dose e termina 14 dias após a última dose da administração das intervenções do estudo para vacinas vivas ou 7 dias no caso de vacinas inativadas*, com exceção da gripe vacinas ou vacina contra a COVID-19 que podem ser consideradas caso a caso.
- Se a vacinação em massa de emergência contra uma ameaça imprevista à saúde pública (por exemplo, uma pandemia) for organizada pelas autoridades de saúde pública fora do programa de imunização de rotina, o período de tempo descrito acima pode ser reduzido se, desde que seja utilizado de acordo com as recomendações governamentais locais e o Patrocinador é notificado.
Sob tais circunstâncias, um participante pode ser considerado elegível para inscrição no estudo e/ou administração de intervenção no estudo após a janela apropriada para atraso ter passado e os critérios de inclusão/exclusão terem sido verificados novamente, e se o participante for confirmado como elegível.
- Administração de medicamentos modificadores da imunidade de ação prolongada a qualquer momento durante o período do estudo.
- Administração de imunoglobulinas e/ou quaisquer produtos sanguíneos ou derivados de plasma desde o nascimento (para bebês com 6 semanas de idade) ou durante o período começando 3 meses antes da administração da primeira dose da(s) intervenção(ões) do estudo ou administração planejada durante o período do estudo.
- Administração crônica (definida como mais de 14 dias no total) de imunossupressores ou outros medicamentos modificadores da imunidade durante o período começando 3 meses antes da(s) primeira(s) dose(s) de intervenção do estudo até o final do estudo. Para corticosteroides, isso significará equivalente de prednisona maior ou igual a (>=) 20 mg/dia para participantes adultos/ >= 0,5 mg/kg/dia com máximo de 20 mg/dia para participantes pediátricos (bebês e crianças). Esteroides inalados e tópicos são permitidos.
Experiência anterior/simultânea em estudos clínicos
• Participar simultaneamente de outro estudo clínico, a qualquer momento durante o período do estudo, no qual o participante foi ou será exposto a uma intervenção investigacional ou não investigacional (vacina, medicamento e dispositivo).
Outras exclusões
- Mulher grávida ou amamentando.
- Mulher que planeja engravidar ou que planeja descontinuar as precauções contraceptivas.
- História/atual consumo crônico de álcool e/ou abuso de drogas. Isso será decidido a critério do investigador.
- Qualquer funcionário do estudo ou seus dependentes imediatos, familiares ou membros da residência.
- Criança sob cuidados.
