Beschreibung
Einschlusskriterien:
- Mindestens 18 Jahre alt.
- Histologisch bestätigter Kopf-Hals-Karzinom mit Plattenepithel-Histologie, der einer lokalen Therapie mit kurativer Absicht (Operation oder Strahlentherapie mit oder ohne Chemotherapie) nicht zugänglich ist.
- Dokumentiertes Behandlungsversagen von mindestens 1 vorangegangener systemischer Therapie in der R/M-Einstellung, sofern nicht als nicht angemessen befunden.
- Messbare Krankheit durch Response Evaluation Criteria in Solid Tumors (RECIST) v1.1.
- Hat einen Tumor, der von HRAS und/oder PIK3CA abhängig ist.
- Leistungsstatus der Eastern Cooperative Oncology Group (ECOG) von 0-1.
- Akzeptable Leber-, Nieren-, endokrine und hämatologische Funktion.
- Muss in der Lage sein, Alpelisib als ganze Tablette oder Suspension zum Einnehmen mit zerkleinerten Tabletten zu schlucken. Für die Verabreichung von Alpelisib darf keine Ernährungssonde verwendet werden.
- Andere protokolldefinierte Einschlusskriterien können gelten.
Ausschlusskriterien:
- Histologisch bestätigte Speicheldrüsen-, Schilddrüsen-, (primäre) kutane Plattenepithel- oder nicht-plattenepitheliale Histologien (z. B. Schleimhautmelanom).
- Laufende Behandlung mit bestimmten Antikrebsmitteln.
- Vorbehandlung (mindestens 1 vollständiger Behandlungszyklus) mit einem FTI- oder PI3K-, mTOR- oder AKT-Inhibitor.
- Behandlung wegen instabiler Angina pectoris, Myokardinfarkt und/oder zerebrovaskulärer Attacke innerhalb der letzten 6 Monate.
- Nicht tolerierbare Neuropathie Grad 2 oder ≥ Grad 3 oder Anzeichen instabiler neurologischer Symptome innerhalb von 4 Wochen nach Tag 1 von Zyklus 1.
- Größere Operation, außer diagnostischer Operation, innerhalb von 2 Wochen vor Zyklus 1, Tag 1, ohne vollständige Genesung.
- Aktive, unkontrollierte bakterielle, virale oder Pilzinfektionen, die eine systemische Therapie erfordern.
- Teilnehmer mit einer gesicherten Diagnose von Diabetes mellitus Typ 1 oder nicht kontrolliertem Typ 2.
- Der Teilnehmer hat eine Beeinträchtigung der Magen-Darm-Funktion (GI) oder eine GI-Erkrankung, die die Absorption der Studienmedikamente nach Ermessen des Prüfarztes erheblich verändern kann.
- Der Teilnehmer hat derzeit eine dokumentierte Pneumonitis/interstitielle Lungenerkrankung.
- Der Teilnehmer hat in der Vorgeschichte schwere Hautreaktionen wie Stevens-Johnson-Syndrom (SJS), Erythema multiforme (EM), toxische epidermale Nekrolyse (TEN) oder Arzneimittelreaktion mit Eosinophilie und systemischen Symptomen (DRESS).
- Andere protokolldefinierte Ausschlusskriterien können gelten.
