A major depressziós rendellenesség (MDD) gyakori, súlyos, visszatérő rendellenesség.
A Seltorexant (JNJ-42847922) a humán orexin-2 receptor (OX2R) hatékony és szelektív antagonistája, amelyet az álmatlanság tüneteivel járó major depressziós rendellenesség (MDDIS) kiegészítő kezelésére fejlesztettek ki.
Ennek a tanulmánynak a hipotézise az, hogy a szeltorexanttal végzett kiegészítő kezelés jobb, mint a placebó a depressziós tünetek kezelésében, a Montgomery Asberg Depresszió Értékelő Skála (MADRS) összpontszámának változásával mérve a kiindulási értéktől a 43. napig az MDDIS-ben szenvedő felnőtt és idős résztvevőknél. nem megfelelő válasz az SSRI/SNRI-kezelésre.

- Teljesítse a Mentális zavarok Diagnosztikai és Statisztikai Kézikönyve – 5. kiadás (DSM-5) diagnosztikai kritériumait a súlyos depressziós rendellenességre (MDD), pszichotikus jellemzők nélkül, a klinikai értékelés alapján, és megerősíti a DSM-5 I. tengelyes rendellenességek klinikai strukturált klinikai interjúja Próbaverzió (SCID-CT), amelyet 60 éves kor előtt diagnosztizáltak az első depressziós epizóddal. The MADRS is a clinician-rated scale designed to measure depression severity and detected changes due to antidepressant intervention.
Scale has 10 items(apparent sadness, reported sadness, inner tension, reduced sleep, reduced appetite, concentration difficulties, lassitude, inability to feel, pessimistic thoughts, and suicidal thoughts) each of which was scored from 0 (not present or normal) to 6 (severe or continuous presence of symptoms).
MADRS total score is sum of scores from individual question items, which ranged from 0 to 60. Higher scores indicates more severe condition.
Negative changes in MADRS total score indicates improvement.
