Visita 1:
Dopo aver esaminato e firmato il consenso, i soggetti verranno sottoposti a screening. Lo screening consisterà in:
Completa IIEF (International Index of Erectile Function) e un questionario sulla frequenza alimentare.
Registrare la storia medica, i farmaci Misurare l'altezza e il peso Vitali Prelevare il sangue per il testosterone totale, la globulina legante gli ormoni sessuali e il TSH (ormone stimolante la tiroide) I soggetti saranno interrogati in merito alla presenza di eruzioni o lesioni genitali
Se il punteggio IIEF è anormale, ai soggetti verrà consigliato di contattare il PCP (fornitore di cure primarie).
I soggetti saranno contattati con i risultati dei test di laboratorio. Se anormale, al soggetto verrà consigliato di contattare il PCP.
Se il soggetto non ha PCP, verranno fornite le informazioni di contatto per un PCP. I soggetti che soddisfano i criteri di inclusione ed esclusione verranno randomizzati nella sequenza uno o nella sequenza due. Una volta arruolati, i soggetti non modificheranno le abitudini alimentari per tutta la durata dello studio se non durante le due sere e i due giorni interi in cui viene fornito il cibo (vedi sotto).
Sequenza uno:
Visita 2:
I soggetti tornano al centro studi la mattina dopo aver digiunato (oltre al consumo di acqua) dalle 23:00 della sera prima. I soggetti non si laveranno i denti né useranno collutori e/o altri rinfrescanti per l'alito quella mattina. Verranno effettuate registrazioni antropomorfiche, della pressione arteriosa e della frequenza cardiaca. Il sangue verrà prelevato e conservato. L'urina sarà raccolta e conservata. Ai soggetti verrà insegnato come utilizzare il dispositivo Rigiscan™. I soggetti riceveranno una cena a base vegetale. I soggetti lasceranno quindi il centro di studio e consumeranno la dieta abituale fino alle 17:00. Per il resto della serata fino alle 23 i soggetti consumeranno solo la cena prevista. L'acqua sarà l'unica bevanda consentita dopo le 17:00. La cena dovrebbe essere consumata alle 20:30, EST (Eastern Standard Time). I soggetti invieranno immagini di tutto ciò che viene consumato (mangiare o bere) dopo le 17:00 al PI oa un rappresentante dello studio un membro del gruppo di studio avrà un contatto testuale o verbale con il soggetto almeno una volta quella sera. I soggetti digiuneranno oltre all'acqua dopo le 23:00. Quella notte, i soggetti eseguiranno una valutazione Rigiscan™ durante la notte a casa. Dovrebbero essere registrate l'ora in cui il soggetto va a letto e l'ora in cui il soggetto si sveglia per la giornata. Il soggetto accetta di stare a letto con Rigiscan™ acceso (tranne che per emergenze o viaggi necessari in bagno) per almeno sette ore.
Visita 3:
I soggetti tornano al centro studi la mattina dopo la valutazione Rigiscan™ e portano il dispositivo Rigiscan™. I soggetti avranno digiunato oltre all'acqua potabile dalle 23:00 della sera prima. I soggetti non si laveranno i denti né useranno collutori e/o altri rinfrescanti per l'alito quella mattina. Verranno effettuate registrazioni antropomorfiche, della pressione arteriosa e della frequenza cardiaca. Il sangue verrà prelevato e conservato. L'urina sarà raccolta e conservata. I soggetti riceveranno quindi un giorno intero di pasti a base vegetale. I soggetti saranno incoraggiati a fare colazione presso il centro studi dopo aver effettuato le misure antropomorfe iniziali, prelevato il sangue e raccolto l'urina. I soggetti invieranno immagini di tutto ciò che viene consumato durante quel giorno al PI e/o a un rappresentante dello studio e un membro del gruppo di studio avrà un contatto testuale o verbale con il soggetto almeno due volte durante quel giorno. La cena dovrebbe essere consumata alle 20:30, EST. I soggetti digiuneranno oltre all'acqua dopo le 23:00. Quella notte, i soggetti eseguiranno una valutazione Rigiscan™ durante la notte a casa. Il caffè nero e/o il tè sono consentiti solo prima di mezzogiorno. Dovrebbero essere registrate l'ora in cui il soggetto va a letto e l'ora in cui il soggetto si sveglia per la giornata. Il soggetto accetta di stare a letto con Rigiscan™ acceso (tranne che per emergenze o viaggi necessari in bagno) per almeno sette ore.
Visita 4:
I soggetti tornano al centro studi la mattina dopo la valutazione Rigiscan™ e portano il dispositivo Rigiscan™. I soggetti non si laveranno i denti né useranno collutori e/o altri rinfrescanti per l'alito quella mattina. Verranno effettuate registrazioni antropomorfiche, della pressione arteriosa e della frequenza cardiaca. Il sangue verrà prelevato e conservato. L'urina sarà raccolta e conservata. Ci sarà quindi un periodo di sospensione per 8-12 giorni e ai soggetti verrà chiesto di tornare alle normali abitudini alimentari durante questo periodo di sospensione.
Visita 5:
Dopo il periodo di washout, i soggetti tornano al centro studi la mattina dopo aver digiunato (oltre al consumo di acqua) dalle 23:00 della sera prima. I soggetti non si laveranno i denti né useranno collutori e/o altri rinfrescanti per l'alito quella mattina. Verranno effettuate registrazioni antropomorfiche, della pressione arteriosa e della frequenza cardiaca. Il sangue verrà prelevato e conservato. L'urina sarà raccolta e conservata. I soggetti riceveranno una cena a base di animali. I soggetti lasceranno quindi il centro di studio e consumeranno la dieta abituale fino alle 17:00. Per il resto della serata fino alle 23 i soggetti consumeranno solo la cena prevista. L'acqua sarà l'unica bevanda consumata dopo le 17:00. La cena dovrebbe essere consumata alle 20:30, EST. I soggetti invieranno immagini di tutto ciò che viene consumato (mangiare o bere) dopo le 17:00 al PI oa un rappresentante dello studio e un membro del gruppo di studio avrà un contatto verbale o testuale con il soggetto almeno una volta quella sera. I soggetti digiuneranno oltre all'acqua dopo le 23:00. Quella notte, i soggetti eseguiranno una valutazione Rigiscan™ durante la notte a casa. Dovrebbero essere registrate l'ora in cui il soggetto va a letto e l'ora in cui il soggetto si sveglia per la giornata. Il soggetto accetta di stare a letto con Rigiscan™ acceso (tranne che per emergenze o viaggi necessari in bagno) per almeno sette ore.
Visita 6:
I soggetti tornano al centro studi la mattina dopo la valutazione Rigiscan™ e portano il dispositivo Rigiscan™. I soggetti non si laveranno i denti né useranno collutori e/o altri rinfrescanti per l'alito quella mattina. Verranno effettuate registrazioni antropomorfiche, della pressione sanguigna e EndoPAT ™. La saliva verrà immagazzinata. Il sangue verrà prelevato e conservato. I soggetti riceveranno quindi un giorno intero di pasti a base di animali. I soggetti saranno incoraggiati a fare colazione presso il centro studi dopo aver effettuato le misure antropomorfe iniziali, aver ottenuto la saliva e prelevato il sangue. I soggetti invieranno immagini di tutto ciò che viene consumato durante quel giorno al PI e/o a un rappresentante dello studio e il team di studio avrà un contatto testuale o verbale con il soggetto almeno due volte durante quel giorno. La cena dovrebbe essere consumata alle 20:30, EST. I soggetti digiuneranno oltre all'acqua dopo le 23:00. Quella notte, i soggetti eseguiranno una valutazione Rigiscan™ durante la notte a casa. Il caffè nero e/o il tè sono consentiti solo prima di mezzogiorno. Dovrebbero essere registrate l'ora in cui il soggetto va a letto e l'ora in cui il soggetto si sveglia per la giornata. I soggetti tornano al centro studi la mattina dopo la valutazione Rigiscan™ e portano il dispositivo Rigiscan™. I soggetti non si laveranno i denti né useranno collutori e/o altri rinfrescanti per l'alito quella mattina. Verranno effettuate registrazioni antropomorfiche, della pressione arteriosa e della frequenza cardiaca. Il sangue verrà prelevato e conservato. L'urina sarà raccolta e conservata.
Sequenza due:
La Sequenza Due è identica alla Sequenza Uno, tranne per il fatto che vengono forniti pasti a base animale invece di pasti a base vegetale. Per i soggetti randomizzati a pasti a base animale nella Sequenza Uno e nella Sequenza Due saranno identici, tranne per il fatto che verranno forniti pasti a base vegetale invece di pasti a base animale.

Criterio di inclusione:
- Il soggetto fornisce il consenso informato scritto e l'autorizzazione HIPAA in inglese prima che venga condotta qualsiasi procedura di studio;
- Nessuna terapia medica (diversa dagli MDI prn (inalatori predosati) e nessun cambiamento negli integratori se applicabile)
- Punteggio IIEF maggiore o uguale a 22
- Il soggetto avrà avuto rapporti sessuali pene-vaginali entro 4 settimane dall'arruolamento. Il punteggio IIEF utilizzato viene convalidato in base a questa finestra temporale.
- Maschio di età compresa tra 18 e 29 anni
- Vive a breve distanza dal Sistema Sanitario Montefiore
- L'altro significativo del soggetto (se applicabile) accetta di supportare il soggetto durante lo studio
- Il soggetto accetta di fotografare e condividere tutti gli oggetti consumati/bevuti durante l'intervento dietetico
- Il soggetto accetta di evitare tutte le droghe illecite, i FANS e l'alcool per almeno due giorni prima della registrazione del Rigiscan™ e nei giorni e nelle notti della registrazione del Rigiscan™
- Il soggetto accetta di astenersi dall'attività sessuale per almeno 36 ore prima del posizionamento del Rigiscan™ e di non avere attività sessuale nei giorni e nelle notti della registrazione del Rigiscan™.
- Il soggetto accetta di non visualizzare, leggere o consumare in altro modo materiale erotico o pornografico per almeno 36 ore prima dei giorni e delle notti della registrazione Rigiscan™ e nei giorni e notti della registrazione Rigiscan™
- Il soggetto accetta di venire a Montefiore per sottoporsi all'addestramento Rigiscan™
- Il soggetto accetta di consumare/bere solo cibi/bevande consentiti.
- Il soggetto accetta di partecipare a tutte le visite di persona a Montefiore, di iniziare il digiuno alle 23:00 la sera prima di ogni visita e di sottoporsi a tutti i prelievi di sangue/salivari/altri test.
- Il soggetto accetta di non usare collutori o collutori o sputare intenzionalmente per l'intera durata dello studio. Ai soggetti verrà chiesto di astenersi dall'usare il collutorio per almeno 2 giorni prima dell'arruolamento. Il soggetto accetta di non lavarsi i denti la mattina del giorno di ciascuna visita di studio. La spazzolatura dei denti è altrimenti consentita.
- BMI inferiore a 30, BMI maggiore o uguale a 18,5, peso superiore a 110 libbre.
- Il soggetto accetta di rispettare le procedure e le visite dello studio.
- Il soggetto si esercita per almeno 15 minuti almeno due volte a settimana. Il soggetto accetta di evitare l'esercizio il giorno prima e il giorno delle misurazioni Rigiscan™
Criteri di esclusione:
- Allergia alimentare rilevante
- Modello dietetico vegetariano o vegano
- Storia di un disturbo alimentare; evidenza di dipendenza da cibo
- Ipertensione e/o storia di ipertensione con o senza trattamento medico
- BMI maggiore o uguale a 30, BMI inferiore a 18,5 o peso inferiore o uguale a 110 libbre
- Malattia medica cronica nota diversa dalla malattia muscoloscheletrica non infiammatoria e dall'asma
- Storia di malattia renale o iperkaliemia
- - Il soggetto ha ricevuto un farmaco sperimentale entro 30 giorni prima della firma del consenso
- Disfunzione erettile
- Ha qualsiasi condizione (ad esempio, malattia psichiatrica) o situazione che, secondo l'opinione dello sperimentatore, può confondere i risultati dello studio o può interferire in modo significativo con la capacità del paziente di aderire alle procedure dello studio
- Attualmente in cura per la malattia di Peyronie
- Livello anormale di testosterone o ormone stimolante la tiroide
- Ipogonadismo trattato o ipotiroidismo
- Viaggio programmato durante lo studio
- Storia di abuso di sostanze negli ultimi 12 mesi
- Uso illecito di droghe, fumo o svapo entro 4 settimane
- Malattia delle vie respiratorie superiori entro due settimane dallo screening
- Se la professione richiede di essere di guardia, nessun lavoro notturno o di guardia durante lo studio
- Qualsiasi pelle trasmissibile o malattia venerea. Il soggetto riferisce eruzioni cutanee o lesioni sul pene o nell'area circostante. Al soggetto verrà chiesto: "hai un'eruzione cutanea o una lesione sul tuo pene o nell'area circostante?"
