ストレスに関連する UCLA/Danone プロバイオティクス介入研究の最適な設計を決定することを目的としたパイロット研究
この研究のパイロット部分 (ステップ 1) の目的は、主観的なストレス反応 (感情)、認知、注意、および生物学的対策(自律神経および代謝反応)。
さまざまな研究対象集団での経験に基づいて、研究者は、健康で均一な集団 (白人、女性、閉経前) が、より高いレベルの状態不安と知覚されたストレスを持ち、実験室のストレス課題 (これは、ステップ 2) のプロバイオティクス介入研究は、ステップ 1 のストレス応答性の根底にある中心的なメカニズムを特定し、次にステップ 2 のプロバイオティクス介入を特定する可能性が最も高くなります。
このパイロット研究では、研究者はベースライン測定を調べて、高ストレスグループと低ストレスグループにおける4つの主観的(感情/認知/注意)ストレスタスク(主要エンドポイント)および生物学的(二次エンドポイント)測定に対する反応の違いを判断します.
調査のステップ 1 の場合、調査員はアンケート データに基づいて高ストレス グループと低ストレス グループへの参加者の層別化を検証し、心理的特性、ラボ ストレス タスク、および潜在的にこれは、最適なカットオフ値、およびステップ 2 の計画されたプロバイオティクス介入研究で実施される最適なストレス課題を決定するのに役立ちます。

- 高知覚ストレスグループの場合: PSS スコア 15 以上および STAI-S 39 以上
- 低知覚ストレス群:PSSスコア8以下、STAI-Sスコア24以下
除外基準:
- 進行中の主要な医学的、心理的、または精神医学的状態、および測定された結果を妨げる向精神薬またはその他の薬の使用における最近の変化(3か月)。
- 現在の神経、心血管、肝臓、腎臓、自己免疫疾患、糖尿病、または癌などの病状。 これには、IBS、IBD、セリアック病、またはその他の栄養欠乏症/疾患、現在の摂食障害、または過去の減量手術を含むがこれらに限定されない GI 障害の現在または過去の 1 年以内の診断が含まれます。
- -不安やうつ病などの心理的状態(つまり、診断および統計マニュアル(DSM-4)の精神医学的診断の履歴がない.
- 事前/併用療法(例えば、PIによって決定されるように、測定された結果を妨げる向精神薬または他の薬物の使用の最近の変化)。
- -前月以内に他の臨床研究に関与した被験者、または別の臨床研究後の除外期間。
- 過去 1 か月間に COVID-19 感染の検査が陽性であった、または過去 2 週間に COVID-19 感染の症状がみられた場合。
ダイエット:
- 過去4週間以内に食生活を変えた参加者。
- プロバイオティクスの参加者は、研究に登録する前に1か月待つように求められます。
- 摂食障害のある参加者。
- -特別な薬用食事(例:肥満、食欲不振、代謝病理学のため)の参加者。
- -過去1か月の人工栄養下の参加者。
- 参加者は、研究の過程で食習慣を変えることを計画しています。
その他の除外事項:
- -妊娠中の女性または研究中に妊娠する予定の女性;スクリーンでのインタビュー(訪問1)およびストレステストの日の尿検査(訪問2)に基づく授乳中の女性。
- ニコチン依存度が中程度から高度の喫煙者 (例: 1 日あたりタバコの 1/2 パック以上)。
- 1日あたり3単位以上のアルコールを定期的に摂取する参加者(1単位= 10mLまたは8g)。
- 理由の如何を問わず、筆記によるアンケート回答ができない参加者。
- 行政上または司法上の決定により、自由を奪われた参加者。
- 法定後見人がいる主要な参加者。
- 抗生物質の参加者は、研究に登録する前に3か月待つように求められます。
承認されていない医薬品:
- アヘン剤/麻薬
- 鎮痛剤の慢性/毎日の使用
- 抗てんかん薬
- 抗うつ薬 (5HT3 や三環系薬剤など) は、3 か月間安定した用量である限り許可されます
- ADD/ADHD の薬 (すなわち、アデロール)
- 食欲抑制のための任意の薬
- 下剤/止瀉薬/胃腸運動に影響を与える薬の慢性的な使用
- インスリン
- 胆汁酸封鎖剤
- -CNS検査を妨げる中枢作用薬の使用
