De effecten van glycine op atherosclerose en metabool syndroom-gerelateerde parameters: een klinische en ex-vivo studie.
De huidige studie zal de centrale hypothese testen dat suppletie met glycine bij mensen het lipidenprofiel verbetert en daardoor het risico op atherosclerose vermindert.
Secundaire uitkomsten, waaronder insulinegevoeligheid en parameters gerelateerd aan het metabool syndroom (MetS) zullen ook worden gemeten.
Bovendien zal een mechanistische studie in een ex-vivo-model de hypothese testen dat glycine via zijn belangrijkste biosynthetische route waarbij serinehydroxymethyltransferase 2 (SHMT2) betrokken is, atherobeschermend is door het remmen van door sterol regulerend element-bindend eiwit 2 (SREBP2) gemedieerde cholesterolbiosynthese in muriene macrofaagachtige cellijn.

Inclusiecriteria:
- Man tussen de 40 en 65 jaar.
- Voldoen aan ten minste drie van de vijf diagnostische criteria voor het metabool syndroom.
- Om hun schriftelijke toestemming te kunnen geven voor deelname aan dit onderzoek.
Uitsluitingscriteria:
- Abnormaal Leverfunctietesten ≥ 3 maal de bovengrens van normaal (ULN).
- Chronische leverziekte anders dan NAFLD.
- Eerdere maag- of dunnedarmoperatie.
- Abnormaal niveau van schildklierstimulerend hormoon (TSH).
- Bekende tabak Meer dan 10 sigaretten per dag roken.
- Bekend alcoholgebruik meer dan 2 drankjes per dag.
- Gebruik van medicijnen waaronder: insuline of insulinesecretagogen, thiazolidinedionen, glucocorticosteroïden, hormoonsubstitutietherapie.
- Koorts > 38,2 °C in de afgelopen 2 weken.
- Auto-immuunziekte of auto-inflammatoire ziekte.
- Chronische nierziekte ≥ stadium III.
- Nefrotisch syndroom.
- Hemoglobine <12 g/dL.
- Metalen clips of implantaten die beeldvorming met magnetische resonantie onmogelijk maken.

Glycine totale dagelijkse dosis van 150 mg/kg driemaal daags verdeeld bij maaltijden (poeder opgelost in 1 kopje water) gedurende 12 weken.
