De wereldwijde doorbloeding van de hersenen neemt af met het vorderen van de leeftijd; sommige volwassenen hebben echter een versnelde afname van de bloedstroom in de hersenen, waardoor ze een groter risico lopen op cognitieve stoornissen. Evenzo neemt de hersenreactiviteit op verhoogde niveaus van koolstofdioxide af met de leeftijd, met een grotere afname bij volwassenen met vasculaire risicofactoren en is verstoord bij de vroege ziekte van Alzheimer. Preklinische modellen suggereren dat een verminderde doorbloeding van de hersenen regionaal resulteert in lage zuurstofniveaus. Momenteel is er een gebrek aan studies bij mensen die de oorzaak of het gevolg van veranderde bloedstroomregulatie in de hersenen onderzoeken.
De onderzoeksdoelen zijn:
- Bepaal de impact van hypoplasie van de wervelslagader (VAH) op de reactiviteit van de hersenen op verhoogde niveaus van koolstofdioxide.
- Bepaal de impact van VAH op de reactie van de bloedstroom in de hersenen op acuut lage zuurstofniveaus.

Inclusiecriteria:
- BMI ≤35 kg/m2
- Niet-roker
- Vrouwelijke proefpersonen: postmenopauzaal
- Momenteel ingeschreven in de Wisconsin Alzheimer's Disease Research Clinical Core
Uitsluitingscriteria:
- Diagnose van milde cognitieve stoornissen of de ziekte van Alzheimer
- Ongecontroleerde hypertensie
- Voorgeschiedenis of bewijs van leverziekte, hematologische ziekte of perifere vasculaire ziekte
- Ernstige nierbeschadiging die hemodialyse vereist
- Hart- en vaatziekten, waaronder: ernstig congestief hartfalen, coronaire hartziekte, ischemische hartziekte (stents, coronaire bypass-transplantaten) en tachycardie
- Geschiedenis van klinisch significante ischemische of hemorragische beroerte, of significante cerebrovasculaire ziekte
- Geschiedenis van hiv/aids
- Ernstige onbehandelde obstructieve slaapapneu
- Geschiedenis van diabetes
- Ernstige neurologische aandoeningen anders dan dementie (bijv. Multiple sclerose, amyotrofische laterale sclerose, hersenchirurgie, enz.)
- Huidige of recente (<1 jaar) ernstige psychiatrische aandoening (As I) of verslavingsstoornissen
- Contra-indicaties voor MRI
