Een fase 1/2 open-label onderzoek om de veiligheid en werkzaamheid van MK-1200 te evalueren bij deelnemers met geavanceerde solide tumoren
Het doel van deze studie is om de werkzaamheid en veiligheid van MK-1200-monotherapie te beoordelen bij deelnemers met gevorderde/gemetastaseerde maag-/gastro-oesofageale junctie (GEJ)-kanker, slokdarmkanker, galwegkanker en ductaal adenocarcinoom van de pancreas die een behandeling met dit middel hebben gehad of daar intolerant voor zijn geweest. tot alle behandelingen waarvan bekend is dat ze klinisch voordeel opleveren.
Deel 1 van het onderzoek zal een dosisescalatie zijn om de maximaal getolereerde dosis (MTD) te bepalen.
Deel 2 evalueert de veiligheid en werkzaamheid van MK-1200 bij 2 verschillende doses

Inclusiecriteria:
- Bevestigde gevorderde (niet-reseceerbare en/of gemetastaseerde) solide tumor: maagkanker (inclusief kanker van de gastro-oesofageale overgang), slokdarmkanker, kanker van de galwegen of ductaal adenocarcinoom van de pancreas
- Deelnemers die bijwerkingen (AE's) hebben ervaren als gevolg van eerdere behandelingen tegen kanker, moeten hersteld zijn tot < graad 1 of baseline
- Bij deelnemers die geïnfecteerd zijn met het humaan immunodeficiëntievirus (HIV) moet het HIV-virus goed onder controle zijn tijdens antiretrovirale therapie
- Hepatitis B-oppervlakteantigeen (HBsAg)-positieve deelnemers komen in aanmerking als ze gedurende ten minste 4 weken antivirale therapie tegen het Hepatitis B-virus (HBV) hebben gekregen en een ondetecteerbare HBV-virale lading hebben
- Deelnemers met een voorgeschiedenis van infectie met het Hepatitis C Virus (HCV) komen in aanmerking als de virale lading van HCV niet detecteerbaar is
- Ontvangen en progressief op of na 1 of 2 eerdere therapielijnen
Uitsluitingscriteria:
- Actieve ernstige spijsverteringsziekte
- Geschiedenis van ernstige hart- en vaatziekten
- Geschiedenis van een acuut hartinfarct; instabiele angina; beroerte of TIA binnen 6 maanden voorafgaand aan de eerste dosis van de onderzoeksinterventie
- Gebruik van een pacemaker
- Diabetes of hypertensie die niet onder controle kan worden gehouden met medicijnen
- HIV-geïnfecteerde deelnemers met een voorgeschiedenis van Kaposi-sarcoom en/of de multicentrische ziekte van Castleman
- Eerder systemische antikankertherapie met onderzoeksmedicatie heeft ontvangen binnen 4 weken vóór de onderzoeksinterventie
- Heeft eerder radiotherapie ontvangen binnen 2 weken na aanvang van de onderzoeksinterventie, of heeft stralingsgerelateerde toxiciteiten, waarvoor corticosteroïden nodig zijn
- Bekende bijkomende maligniteit die verergert of die in de afgelopen twee jaar een actieve behandeling vereist heeft
- Bekende actieve metastasen van het centrale zenuwstelsel (CZS) en/of carcinomateuze meningitis
- Actieve infectie die systemische therapie vereist
- U bent niet voldoende hersteld van een grote operatie of u heeft aanhoudende chirurgische complicaties

Experimenteel: Deel 1: MK-1200
In deel 1 ontvangen de deelnemers elke 2 weken (Q2W) oplopende doses MK-1200 via intraveneuze (IV) infusie totdat aan eventuele stopzettingscriteria is voldaan.
