Langetermijnverlengingsonderzoek (AtDvance) om de veiligheid en werkzaamheid van GSK1070806 te evalueren bij deelnemers met matige tot ernstige atopische dermatitis.

Inclusiecriteria:

  • Deelnemers moeten het toestemmingsdocument ondertekenen en dateren.
  • Deelnemers met de diagnose matige tot ernstige atopische dermatitis (AtD) die de kwalificerende fase 2-ouderstudie (NCT05999799) van GlaxoSmithKline (GSK's) AtD hebben voltooid en die naar hun mening baat kunnen hebben bij GSK1070806.
  • Wees opzettelijk en in staat om op afspraak de arts in de kliniek te bezoeken en volg alle procedures met betrekking tot onderzoeken en vragenlijsten (in staat om door de patiënt gerapporteerde uitkomsten (PRO) vragenlijsten te lezen en te begrijpen en om elektronische apparaten te kunnen gebruiken).
  • Het gebruik van anticonceptiemethoden voor vrouwen moet in overeenstemming zijn met de lokale regelgeving inzake anticonceptiemethoden van deelnemers aan klinische onderzoeksprogramma's.
  • Vrouwelijke deelnemers komen in aanmerking voor deelname aan het onderzoeksprogramma als zij niet zwanger zijn of borstvoeding geven en aan een van de volgende voorwaarden voldoen:
  • Het is Vrouw die niet zwanger kan worden (WONCBP).
  • Het is een vrouw die zwanger kan worden (WOCBP) en gebruikt een zeer effectieve anticonceptiemethode met een faalpercentage van minder dan (<) 1 procent (%) tijdens de proefdosisperiode en gedurende ten minste 16 weken na de laatste dosis van het onderzoeksgeneesmiddel . We moeten de waarschijnlijkheid van falen van de anticonceptie (bijvoorbeeld niet-meewerken, vroege anticonceptie) in verband met de eerste dosis van het medicijn beoordelen.
  • WOCBP moet een negatief resultaat behalen in een zeer gevoelige zwangerschapstest (via urine- of serumtest, zoals voorgeschreven door lokale regelgeving) voordat de eerste dosis van het onderzoeksgeneesmiddel wordt toegediend.
  • Als de urinetest positief is of als het negatieve resultaat niet kan worden bevestigd (dat wil zeggen als het resultaat onduidelijk is), is een zwangerschapstest door middel van een serumtest vereist. In dergelijke gevallen is het testresultaat positief als het serum wordt getest. Deelnemers moeten worden uitgesloten van het onderzoeksproject.
  • Aanvullende vereisten voor het testen van de zwangerschap tijdens en na blootstelling aan het medicijn.
  • De onderzoeker is verantwoordelijk voor het onderzoeken van de medische geschiedenis, de geschiedenis van de menstruatiecyclus en de seksuele activiteit op de korte termijn. Om het risico te verminderen van het screenen van zwangere vrouwen die mogelijk niet worden gedetecteerd aan het begin van de zwangerschap.

Uitsluitingscriteria:

  • Definitieve stopzetting van het onderzoeksgeneesmiddel op elk moment tijdens GSK's kwalificerende Fase 2 AtD (NCT05999799) of een medische aandoening die de deelname van GSK aan het Fase 2 219538 (NCT05999799) AtD-onderzoeksproject zou belemmeren.
  • Deelnemers die tijdens het kwalificerende Fase 2 (NCT05999799) AtD-onderzoeksproject van GSK een bijwerking (AE) of een ernstige bijwerking (SAE) ontwikkelden op basis van laboratoriumparameters, lichamelijk onderzoek, vitale functies, elektrocardiogram (ECG), medische geschiedenis, enz. De geschiedenis wijst er volgens de onderzoekers op dat als het onderzoeksgeneesmiddel (IMP) wordt voortgezet, dit onnodige risico's voor de deelnemers kan veroorzaken.
  • Topische medicijnen voor AtD binnen 1 week voorafgaand aan uw afspraak op dag 1, zoals: Topische calcineurineremmers (TCI)/topische corticosteroïden (TCS), fosfodiësterase-4 (PDE-4) en Janus-activeringskinaseremmers (JAKi) voor uitwendig gebruik .
  • Topische corticosteroïden (TCS) (zoals hydrocortison, betamethason)
  • Topische calcineurineremmers (TCI) (zoals tacrolimus, pimecrolimus)
  • Fosfodiësterase-4 (PDE4)-remmer voor uitwendig gebruik (bijv. Crisaborol)
  • JAKi voor extern gebruik (bijv. ruxolitinib)
  • Medicijnen voor plaatselijk gebruik, of andere kruiden-/traditionele medicijnen die de AtD waarin de deelnemers zich bevinden, kunnen beïnvloeden.
  • Deelnemers die systemische therapie kregen, die als gecontra-indiceerd wordt beschouwd, inclusief systemische therapie gebruikt als noodmedicatie voor AtD, vanaf de screening voor GSK's Fase 2 AtD 219538 onderzoeksproject tot het LTE-protocol begon, konden niet deelnemen aan het onderzoeksproject.

Beschrijving IPD-plan

Gekwalificeerde onderzoekers kunnen via het Data Sharing Portal toegang vragen tot geanonimiseerde individuele gegevens op patiëntniveau (IPD) en gerelateerde onderzoeksdocumenten van de in aanmerking komende onderzoeken. IPD-tijdsbestek voor delen

Geanonimiseerde IPD zal beschikbaar worden gesteld binnen zes maanden na publicatie van primaire, belangrijke secundaire en veiligheidsresultaten voor onderzoeken naar producten met goedgekeurde indicatie(s) of beëindigde middelen voor alle indicaties.

IPD-toegangscriteria voor delen

Geanonimiseerde IPD wordt gedeeld met onderzoekers wier voorstellen zijn goedgekeurd door een onafhankelijk beoordelingspanel en nadat er een overeenkomst voor het delen van gegevens is gesloten. Toegang wordt verleend voor een initiële periode van twaalf maanden, maar indien gerechtvaardigd kan een verlenging worden verleend tot maximaal zes maanden.
