Een vervolgonderzoek naar TAK-279 bij volwassenen met colitis ulcerosa (UC) en de ziekte van Crohn (CD)
Een fase 2, open-label extensieonderzoek om de veiligheid en verdraagbaarheid op lange termijn van orale zasocitinib (TAK-279) te evalueren bij deelnemers met matig tot ernstig actieve colitis ulcerosa en matig tot ernstig actieve ziekte van Crohn
De ziekte van Crohn en colitis ulcerosa zijn twee soorten inflammatoire darmziekten (IBD), een ernstige, langdurige aandoening in de darmen die pijn en zwelling (ontsteking) in de darm kan veroorzaken. TAK-279 is een geneesmiddel dat ontstekingen helpt blokkeren.
Deze studie is een uitbreiding van de ouderstudies, TAK-279-CD-2001 (NCT06233461) en TAK-279-UC-2001 (NCT06254950). Dit betekent dat deelnemers die in een van de ouderstudies reageerden op de behandeling met TAK-279 mogelijk kunnen blijven profiteren van de behandeling in deze studie.
Het belangrijkste doel van deze studie is om erachter te komen hoe veilig TAK-279 is voor langdurig gebruik en om na te gaan of het de darmontsteking en -symptomen vermindert bij langdurig gebruik bij volwassenen met matig tot ernstig actieve UC of CD.
De deelnemers worden maximaal 2 jaar (108 weken) behandeld met TAK-279.
Tijdens het onderzoek bezoeken de deelnemers hun onderzoekskliniek 11 keer.

Inclusiecriteria:
De deelnemer is bereid en in staat om de procesprocedures en vereisten (inclusief digitale hulpmiddelen en toepassingen) te begrijpen en volledig na te leven, naar het oordeel van de onderzoeker.
De deelnemer heeft geïnformeerde toestemming gegeven (dat wil zeggen schriftelijk, gedocumenteerd via een ondertekend en gedateerd formulier voor geïnformeerde toestemming [ICF]) en alle vereiste privacy-autorisatie voorafgaand aan de start van eventuele procesprocedures.
Ziektespecifieke inclusiecriteria:
- Voltooiing van week 52 in de ouderfase 2 CD- en UC-onderzoeken met geldige elektronische (e) dagboekgegevens voor week 52 (TAK-279-CD-2001 en TAK-279-UC-2001).
Klinische of symptomatische responder bij ouderonderzoek in week 52, zoals hieronder gedefinieerd:
- TAK-279-CD-2001: Klinische respons in PRO2 tijdens week 52 van het ouderonderzoek, beoordeeld als >=30% afname van de gemiddelde dagelijkse zeer zachte of vloeibare ontlasting en/of >=30% afname van de gemiddelde AP ten opzichte van de uitgangswaarde van het ouderonderzoek.
- TAK-279-UC-2001: Symptomatische respons in week 52 van het ouderonderzoek, beoordeeld als een reductie in de gedeeltelijk gemodificeerde Mayo-score (pmMS) van >=1 punten en >=30% ten opzichte van de uitgangswaarde van het ouderonderzoek; en een afname ten opzichte van de uitgangswaarde van het ouderonderzoek in de subscore voor rectale bloedingen van >=1 punt of een absolute subscore voor rectale bloedingen van <=1 punt.
Andere algemene inclusiecriteria:
- Deelnemers moeten voldoen aan de anticonceptie-aanbevelingen.
Uitsluitingscriteria:
- Deelnemer die door de onderzoeker als ongeschikt wordt beschouwd voor het OLE-onderzoek vanwege zijn of haar zorgen over de naleving van het onderzoek en de therapietrouw.
Deelnemers met maligniteit of dysplasie per endoscopie op enig moment tijdens de ouderstudie of aan het begin van de OLE.
Uitsluitingscriteria met betrekking tot laboratoriumonderzoek:
Deelnemers die voldoen aan de uitsluitingscriteria met betrekking tot laboratoriumonderzoek zoals gedefinieerd in het protocol.
Uitsluitingscriteria met betrekking tot andere verboden gelijktijdige medicatie:
- Deelnemers die orale corticosteroïden gebruikten voor CD of UC tijdens de ouderstudie in of na week 48.

The responses to the 13 items on the FACIT-Fatigue questionnaire are each measured on a 5-point Likert scale, where 0=Not at all, 1=A little bit, 2=Somewhat, 3=Quite a bit and 4=Very much.
The total score ranges from 0 to 52.
High scores represent less fatigue.
