Un estudio de continuación de TAK-279 en adultos con colitis ulcerosa (CU) y enfermedad de Crohn (EC)
Un ensayo de extensión de etiqueta abierta de fase 2 para evaluar la seguridad y tolerabilidad a largo plazo del zasocitinib oral (TAK-279) en participantes con colitis ulcerosa activa de moderada a grave y enfermedad de Crohn activa de moderada a grave
La enfermedad de Crohn y la colitis ulcerosa son dos tipos de enfermedad inflamatoria intestinal (EII), que es una afección grave y prolongada en el intestino que puede causar dolor e hinchazón (inflamación) en el intestino. TAK-279 es un medicamento que ayuda a bloquear la inflamación.
Este estudio es una extensión de los estudios principales, TAK-279-CD-2001 (NCT06233461) y TAK-279-UC-2001 (NCT06254950). Esto significa que los participantes que respondieron al tratamiento con TAK-279 en cualquiera de los estudios principales pueden continuar beneficiándose del tratamiento en este estudio.
El objetivo principal de este estudio es descubrir qué tan seguro es TAK-279 para el uso a largo plazo y verificar si reduce la inflamación y los síntomas intestinales cuando se usa durante un período más prolongado en adultos con CU o EC activa de moderada a grave.
Los participantes serán tratados con TAK-279 por hasta 2 años (108 semanas).
Durante el estudio, los participantes visitarán la clínica del estudio 11 veces.

Descripción
Criterios de inclusión:
El participante está dispuesto y es capaz de comprender y cumplir plenamente con los procedimientos y requisitos del ensayo (incluidas las herramientas y aplicaciones digitales), en opinión del investigador.
El participante ha proporcionado su consentimiento informado (es decir, por escrito, documentado mediante un formulario de consentimiento informado [ICF] firmado y fechado) y cualquier autorización de privacidad requerida antes del inicio de cualquier procedimiento del ensayo.
Criterios de inclusión específicos de la enfermedad:
- Finalización de la semana 52 en los ensayos de CD y UC de fase 2 principales con datos del diario electrónico (e) válidos para la semana 52 (TAK-279-CD-2001 y TAK-279-UC-2001).
Respondedor clínico o sintomático en la semana 52 del ensayo principal, como se define a continuación:
- TAK-279-CD-2001: Respuesta clínica en PRO2 en la semana 52 del ensayo principal, evaluada como una disminución >=30 % en el promedio diario de deposiciones muy blandas o líquidas y/o una disminución >=30 % en el AP promedio desde el inicio del ensayo principal.
- TAK-279-UC-2001: Respuesta sintomática en la semana 52 del ensayo principal, evaluada como una reducción en la puntuación Mayo modificada parcial (pmMS) de >=1 punto y >=30 % con respecto al valor inicial del ensayo principal; y una disminución con respecto al valor inicial del ensayo principal en la subpuntuación de sangrado rectal de >=1 punto o una subpuntuación absoluta de sangrado rectal de <=1 punto.
Otros criterios generales de inclusión:
- Los participantes deben cumplir con las recomendaciones de anticoncepción.
Criterios de exclusión:
- Participante considerado por el investigador como inadecuado para el ensayo OLE debido a sus preocupaciones sobre el cumplimiento del ensayo y la adherencia a la medicación.
Participantes con neoplasia maligna o displasia por endoscopia en cualquier momento durante el ensayo principal o al comienzo del OLE.
Criterios de exclusión relacionados con investigaciones de laboratorio:
Participantes que cumplan con los criterios de exclusión relacionados con investigaciones de laboratorio según lo definido en el protocolo.
Criterios de exclusión relacionados con otros medicamentos concomitantes prohibidos:
- Participantes que tomaron corticosteroides orales para la EC o la CU durante el ensayo principal en la semana 48 o después.

The total score ranges from 0 to 52.
High scores represent less fatigue.
