Pilotfase I/II-studie av aminosyreleucin i behandling av pasienter med transfusjonsavhengig Diamond Blackfan-anemi (LeucineDBA)
Bruken av nye terapier for å rekonstituere blodcelleproduksjon og fremme organytelse ved bruk av benmargssvikt som modell: en pilotfase I/II-studie av aminosyreleucin i behandling av pasienter med transfusjonsavhengig diamant-blackfan-anemi
Denne studien vil fastslå sikkerheten og muligheten for å gi aminosyren leucin til pasienter med Diamond Blackfan-anemi (DBA) som er avhengige av transfusjoner av røde blodlegemer.
Leucinet forventes å gi en respons hos pasienter med DBA til et punkt hvor produksjonen av røde blodlegemer øker. Transfusjoner av røde blodlegemer kan da være sjeldnere eller muligens seponeres.
Etterforskerne vil studere eventuelle bivirkninger ved å gi leucin til DBA-pasienter. Leucinnivåer av leucin vil bli oppnådd ved baseline og under studien.
Legemidlet leucin vil bli gitt i kapselform og tatt 3 ganger om dagen i totalt 9 måneder.

Leucin vil bli gitt til deltakerne i form av en kapsel og vil bli tatt tre ganger daglig.
Blodhemoglobinnivået vil bli overvåket hver 3-4 uke i 9 måneder.
Hele studiet vil vare i 12-15 måneder.
Forsøkspersonene må være to år eller eldre og på transfusjon i mer enn seks måneder før påmelding.

Inklusjonskriterier:
- diagnostisert med Diamond Blackfan-anemi som publisert i British Journal of Hematology
- transfusjonsavhengig
- alder 2 år og eldre
- tilstrekkelig nyrefunksjon
- tilstrekkelig leverfunksjon
- negativ B-HCG dersom pasienten er en menstruerende kvinne og dokumentasjon på adekvat prevensjon
- undertegnet informert samtykke
Ekskluderingskriterier:
- Kjent overfølsomhet overfor forgrenede aminosyrer
- Diagnose av en medfødt feil ved aminosyremetabolismeforstyrrelse
- Tidligere hematopoetisk stamcelletransplantasjon
- Graviditet, eller planlegger å bli gravid i løpet av prøveperioden
